Bcl-2基因对栗疫病菌细胞凋亡的影响及其转化子菌株的表型测定

【摘要】 Bcl-2家族蛋白是目前已知的细胞凋亡中最重要的调控因子,在细胞凋亡通路中起着重要的调节作用。Bcl-2蛋白家族有两种成员:抗凋亡蛋白和促凋亡蛋白,并在线粒体水平上决定细胞的存活或死亡,动物细胞中Bcl-2家族的蛋白能够通过调节线粒体中细胞色素-C的释放来调节细胞凋亡。本文主要通过把外源bcl-2基因转入栗疫病菌中,采用RT-PCR的方法检测bcl-2基因在栗疫病菌株中的表达情况,对表达了bcl-2基因的转化子及野生型的菌株进行了病毒传递率和对国光苹果致病性的测定,结果表明表达了bcl-2基因的转化子表现出病毒传递率提高、致病性增强等一系列特征。由此可知bcl-2基因对栗疫病菌细胞凋亡起调节作用,并对病菌的生物学特征有一定影响。Bcl-2家族的蛋白在丝状真菌的整个生长发育过程中调节其生长及形态学变化。对表达6ax基因的菌株能表现出对寄主不致病,并伴随着细胞凋亡的特征;对表达bcl-2基因的菌株能够表现出对外界压力抗性的增强,对寄主的致病性增强的特征。本研究通过对转入bcl-2基因的3个转化子菌株及野生型菌株在各种胁迫培养基上生长速率的测定,检测表达bcl-2基因的栗疫病菌株能否表现出对外界各种压力抗性增强的特征,结果表明表达bcl-2基因的3个转化子菌株在含高盐、氧化剂、高渗培养基上的生长速率明显高于野生型菌株。这样的结果表明,bcl-2基因能够提高栗疫病菌株抵抗外界环境的胁迫压力。

【Abstract】 Proteins belonging to the Bcl-2 family are key regulators of apoptotic cell death.Bcl-2 family has two different members:anti-apoptotic protein and pro-apoptotic protein, and they regulate apoptosis in animals by controlling mitochondria efflux of cytochrome c.Here, we transformed bcl-2 gene into protoplast of Cryphonectria parasitic and tested the expression of bcl-2 gene. We tested virus transportation rate of transformants and wt-strains,we also tested their hyper virulent to host.The results indicated that strains expressing the anti-apoptotic Bcl-2 protein can enhance virus transportation rate and are hyper virulent to host.Thus bcl-2 gene also played a regulatory role in apoptosis of Cryphonectria parasitica and had some infection in the biological characters.Proteins belonging to the Bcl-2 family are tightly linked with growth and morphogenetic adaptation that occur throughout the entire fungal life cycle.Isolates expressing the pro-apoptotic bax gene underwent cell death with apoptotic characteristics and showed non-pathogenic life styles.Isolates expressing the anti-apoptotic bcl-2 gene exhibited enhanced stress resistance and were hyper virulent to host plants.Here we tested the growth rate of transformants and wt-strains on various threatening culture mediums in order to know if the transformants of Cryphonectria parasitica showed enhanced stress resistance.The results indicated that the transformants had faster growth rate than wt-strains on various threatening culture mediums.Thus bcl-2 gene can enhance stress resistance in Cryphonectria parasitica.
