从风论治过敏性哮喘的文献整理和实验研究

【摘要】 过敏性哮喘属于Ⅰ型变态反应性疾病,是机体受到抗原刺激后产生特异性免疫应答而引起组织损伤和/或生理功能紊乱的一种疾病,是临床常见病、多发病。该病散在于传统医学的多种疾病当中,也可表现为独立疾病。主要是外源性变态反应原引起的致组织损伤和生理功能紊乱。我国的过敏性哮喘发病率呈上升趋势,严重影响人们的身心健康。治疗和预防过敏性哮喘的发生具有重要的意义。此研究不仅为临床防治过敏性哮喘提供一定思路,而且也为中西医结合防治过敏性哮喘提供实验依据和新的途径。1目的通过文献研究确认风邪与过敏性哮喘病因病机及发病、治疗的相关性;通过实验研究获得以祛风药为君药的加味过敏煎防治过敏性哮喘的实验数据,阐释其抗过敏机制,为从风邪论治过敏性哮喘提供理论指导和实验依据。2方法2.1文献研究本研究采用文献资料法、逻辑分析法和统计学方法,对历代文献关于风邪致病以及过敏性哮喘发病相关文献进行梳理,探讨历代医家对风邪与过敏性哮喘病因病机及发病、治疗的相关性认识。(1)分析历代文献中哮喘的分类,筛选出属于过敏性哮喘的文献记载。(2)研究过敏性哮病因病机(3)研究过敏性哮喘发病症状(4)收集、整理古代文献中治疗哮喘的方剂,重点研究属于治疗过敏性哮喘的药物使用概率。2.2实验研究2.2.1实验动物清洁级SD大鼠,雄性36只,体重180—200g,由北京维通利华公司提供。随机分为正常对照组、哮喘模型组、加味过敏煎给药组,每组各12只。除正常对照组外,其他各组大鼠每只隔天腹腔注射1ml抗原液(含有卵蛋白100mg、氢氧化铝100mg、百日咳杆菌疫苗5×109个)致敏。正常对照组腹腔注射等量生理盐水。致敏14d后,给药治疗。给药组给予加味过敏煎治疗,正常对照组和哮喘模型组给予等量生理盐水。给药1h后,以卵蛋白雾化吸入激发哮喘,给药和激发连续14d。于末次激发后1小时开始试验取材。饲养条件:自然光照,室温(冬春季室温为17℃±2℃,夏秋季室温为25℃±2℃)。自由摄取水及饲料,饲料为普通鼠全价颗粒饲料(北京实验动物研究中心提供)。2.2.2检测方法血清免疫球蛋白E(IgE)含量测定采用酶联免疫吸附实验(ELISA)进行测定。2.2.3实验结果统计学分析方法用SPSS13.0统计分析软件处理数据。各组数据均采用均数±标准差(x±s)表示(P<0.05为差异显著,P<0.01为差异十分显著)。3结果3.1文献研究结果风邪与过敏性哮喘病因病机及发病、治疗具有密切关系。3.2实验研究结果哮喘模型组血清IgE水平与正常对照组相比显著升高(P<0.01)。加味过敏煎给药组可显著降低IgE水平(P<0.05),与哮喘模型组相比具有显著差异。本实验研究结果显示,哮喘模型组动物血清中IgE水平显著升高,具备Ⅰ型变态反应特征。4结论4.1过敏性哮喘的发生与风邪的侵袭密切相关本研究采用文献资料法、逻辑分析法和统计学方法,对历代文献关于风邪致病以及过敏性哮喘发病相关文献进行梳理研究,认识到风邪与过敏性哮喘病因病机及发病、治疗具有密切关系。4.2加味过敏煎具有治疗过敏性哮喘的作用实验观察到加味过敏煎可以降低IgE水平,抑制哮喘过敏性炎症反应,表明过敏煎具有治疗过敏性哮喘的作用,其作用机理可能与抗炎性渗出、抗变态反应有关,其进一步的机制仍有待深入探讨。

【Abstract】 Allergic asthma are clinical common and frequently-occurring I-allergic disease, which is the body created by the antigen-specific immune response after stimulation caused by tissue damage and/or physiological dysfunction of a disease. The disease is scattered among a number of diseases in traditional medicine, can also be expressed as an independent disease, which is mainly caused by exogenous allergen-induced tissue damage and physiological dysfunction. Our incidence of allergic asthma on the rise, seriously affecting people’s physical and mental health. Therefore, treatment and prevention of the occurrence of allergic asthma is of great significance. This study not only for clinical prevention and treatment of allergic asthma to provide some ideas, but also for Chinese and Western medicine with prevention and treatment of allergic asthma to provide experimental evidence and a new way.lObjectiveThrough Theoretical studies have confirmed pathogenic wind and allergic asthma etiology and pathogenesis and the incidence of illness, treatment-related; Obtained by experimental study with Supplemented Guo Min Jian prevention and treatment of allergic asthma fried experimental data to explain its anti-allergic mechanisms, from the pathogenic wind treating Allergic Asthma provide theoretical and experimental basis for the guidance.2 Methods2.1 Theoretical methodsIn this study, literature and data, logic analysis and statistical methods, pathogenic wind pathogenicity as well as the ancient literature on the pathogenesis of allergic asthma in the literature should be straightened out to explore the ancient physicians of pathogenic wind and allergic asthma machine and the incidence of illness, treatment understanding of the relevance.(1) Analysis of ancient literature, the classification of asthma, allergic asthma screening that belongs to the documentary record.(2) Research Etiology and Pathogenesis of Allergic Asthma(3) studies of allergic asthma symptoms (4) Analysis of various types of asthma the frequency of drug use, focusing on part of the treatment of allergic asthma probability of drug use.2.2 Experimental methods2.2.1 Experimental animalsThirty-six Sprague Dawley male rats, weighted 180-260g, brought from Beijing Weitong Lihua animal study center, Were randomly divided into normal control group, asthma model group, fried treatment group with Supplemented Guo Min Jian,12 rats in each group. Apart from the normal control group, rats in each group every two days after intraperitoneal injection of lml antigen solution (containing egg protein100mg, aluminum hydroxide 100mg, Bordetella pertussis vaccine 5×109piece) allergen. The normal control group, intraperitoneal injection of equivalent normal saline.14d sensitization after drug treatment with Supplemented Guo Min Jian, The normal control group and the asthma model group were given equivalent normal saline, 1h after the neb nebulised medication in order to stimulate ovalbumin inhalation asthma, drug delivery and stimulate continuous 14d. lhour after the last excitation started experimenting with drawn. All rats received natural light, ate and drank freely, and were controlled under certain room temperature was 17℃±2℃in winter and spring, and it was 25℃±2℃in summer and autumn.2.2.2 Test methodTo assay the contents of IgE, ELISA was engaged.2.2.3 Analysis methodThe differences of groups were tested by SPSS 13.0. All the data were showed by mean±standard error. P<0.05 meant obvious difference. P<0.01 meant very obvious difference.3Results3.1 Results of theoretical studypathogenic wind and allergic asthma mechanism and the incidence of illness treatment with closely related.3.2 Results of experimentsSerum IgE levels of asthma model group was significantly higher compared to normal control group(P<0.01). Treatment group with Supplemented Guo Min Jian can significantly reduce the IgE levels (P<0.05), compared with the asthma model group with significant difference. This study showed that asthma model group animals’s serum IgE level was significantly increased with I-type allergic reaction characteristics. 4 Conclusions4.1 The occurrence of allergic asthma is closely related to the invasion of pathogenic wind. In this study, literature and data, logic analysis and statistical methods, pathogenic wind pathogenicity as well as the ancient literature on the pathogenesis of allergic asthma combing literature research, recognizing pathogenic wind and allergic asthma machine and the incidence of illness treatment with close relationship.4.2Supplemented Guo Min Jian could therapy allergic asthma. Experimental observation that Supplemented Guo Min Jian can reduce the levels of IgE, inhibit allergic inflammation in asthma, indicating that Supplemented Guo Min Jian could treate the allergic asthma, and its mechanism of action may be related to anti-inflammatory exudation, anti-allergy and its further-depth study of the mechanism remains to be femtocell.
