骨形态发生蛋白2和血管内皮生长因子基因在大鼠股骨骨不连中不同区域的动态表达

【摘要】 目的观察大鼠股骨骨不连修复过程中骨缺损区不同部位骨形态发生蛋白-2(bone morphogenetic protein-2, BMP-2)与血管内皮生长因子(Vascular endothelial growth factor, VEGF)基因在不同时间段的表达情况。方法SD大鼠建立股骨骨不连模型,分别于造模后1,3,7,14,21,28,35d,各取5只大鼠,处死取材,RT-PCR技术检测大鼠股骨骨形态发生蛋白2和血管内皮生长因子mRNA的表达。结果大鼠股骨在骨不连修复过程的不同时间,BMP-2和VEGF的基因表达在中心区和外周区存在差异,中心区BMP-2和VEGF基因表达水平的增幅及峰值低于边缘区,且表达时间滞后。结论大鼠股骨骨不连缺损中心区域在修复早期BMP-2和VEGF基因的低表达及时间滞后可能是其骨不连发生的重要原因,在相应的时期补充外源性BMP-2和VEGF可能助于骨不连的修复愈合。

【Abstract】 ObjectivesTo observe the dynamic expression of bone morphogenetic protein-2(BMP-2) and vascular endothelial growth factor(VEGF)gene during restoration of rat femoral nonunion.MethodThe models of femoral nonunion of SD rats were established, and then the genic expression of BMP-2 and VEGF in different repairing stage of nonunion(day 1,3,7,14,21,28,35)were detected using RT-PCT technique.ResultsIn different stages of femoral nonunion, the genic expression of BMP-2 and VEGF gene between central zone and outer zone also varied, their genic expression increment and the peak level were significantly lower In the central zone than those in the outer zone, and the peak level in the central zone appeared later than that of in the outer zone.ConclusionLower value and the lag in genic expression of BMP-2 and VEGF in the central zone during the early stage may be an important reason of nonunion occurring in rat femur, supplying extrinsic source of BMP-2 and VEGF in early stage may be contribute to the repairing of nonunion.
