穿心莲内酯对小鼠呼吸道非特异性免疫功能的影响

【摘要】 目的:研究穿心莲内酯对呼吸道非特异性免疫功能的影响,探索穿心莲内酯治疗呼吸系统疾病新的作用机制。方法:用鼠伤寒沙门氏菌菌液对小鼠进行滴鼻感染,造出细菌性肺炎动物模型。将此肺炎模型小鼠随机分为模型组、穿心莲内酯高、低剂量治疗组,并设立正常对照组。对模型组和正常对照组小鼠,用生理盐水(0.5ml/只)腹腔注射;对穿心莲内酯高、低剂量组小鼠分别按200mg/kg、100mg/kg进行腹腔注射给药。自由饮水和采食,一周后,在最后一次给药,24h后。用戊巴比妥钠腹腔注射麻醉;进行小鼠眼球摘除采血,血样收至含有肝素钠的收集管中供血常规测定;脱颈处死小鼠,打开气管用生理盐水进行灌注,收集支气管肺泡灌洗液(BALF)。打开胸腔和腹腔,取肺脏、脾脏和胸腺,测定各组小鼠肺脏,脾脏和胸腺指数;显微镜下观察HE染色肺组织的病理变化、计算BALF中单核巨噬细胞吞噬功能和T淋巴细胞阳性百分率;比浊法测定BALF中溶菌酶的活性;用小鼠双抗体夹心ABC-ELISA试剂盒测定BALF中分泌型免疫球蛋白A和干扰素-γ的含量。结果:与正常对照组相比,细菌性肺炎模型组小鼠肺脏、胸腺及脾脏指数显著升高(p<0.01);其中肺脏发生组织形态学变化,主要为炎性细胞浸润,毛细血管充血,肺泡壁增厚,纤维蛋白渗出等;血液中白细胞总数(WBC)淋巴细胞数量(LYM)、单核细胞数量(MON)和中性粒细胞数量(GRA)都有显著升高(p<0.05);支气管肺泡灌洗液中肺泡巨噬细胞(AM)吞噬功能显著升高(p<0.05);分泌型免疫球蛋白A(sIgA)和干扰素-γ(INF-γ)含量显著升高(p<0.01);而BALF中溶菌酶的含量显著降低(p<0.01)。与模型组比较,穿心莲内酯治疗组小鼠肺组织的病理变化得到缓解改善,炎症明显减轻;肺脏、脾脏和胸腺指数有显著降低(p<0.05);脾脏和胸腺增生得到有效控制;BALF中AM的吞噬功能和溶菌酶的含量得到显著提高(p<0.01);BALF中sIgA和IFN-γ含量基本降低到正常水平。结论:穿心莲内酯可缓解鼠伤寒沙门氏菌诱导的小鼠肺部炎症,其作用机制可能与增强巨噬细胞吞噬功能和溶菌酶活性;改善肺部病变,缓解炎症;抑制细菌感染诱导的免疫器官增生;间接地使BALF中sIgA分泌及IFN-γ表达降低并接近正常水平相关。

【Abstract】 Objective:To investigate the effect of andrographolide on the respiratoty tract nonspecific immune functions.Methods:The mice model of pneumonia were built up. The pneumonia mice were treated with different concentrations of andrographolide.A week later,the thmus index,the spleen index,the phagocytosis of macrophages and the contents of secretory immunoglobulin A(sIgA) and interferon-γ(IFN-γ) in the bronchoalveolar lavage fluid(BALF) of pneumonia mice were tested.Results: Compared with the nomal group,the thmus index,the spleen index,and the contents of secretory immunoglobulin A(sIgA) and interferon-γ(IFN-γ) in the model group increased obviously,the contents of lysozyme decreased in the BALF.Compared with the model group,the thmus index,the spleen index,and the contents of secretory immunoglobulin A(sIgA) and interferon-γ(IFN-γ) in the BALF of high dose andrographolide group decreased obviously,the phagocytosis of macrophages in the BALF of high dose andrographolide group increased significantly.Conclusion:The mechanism of andrographolide is complex,andrographolide has the enhancement on the phagocytosis of macrophages and the contents of lysozyme,has the inhibitory action on immunity organs,the secretion of sIgA and the content of IFN-γ.
