高等真菌Sarcodon joedes子实体化学成分的研究

【摘要】 高等真菌Sarcodon joedes为担子菌纲真菌,主要分布于我国云南省。近年来国内外学者从Sarcodon属高等真菌中发现了许多结构新颖、生物活性多样的次生代谢产物,,但Sarcodon joedes的化学成分和生物活性的研究尚未见报道。本文以采自云南昆明的高等真菌Sarcodon joedes的子实体为研究对象,利用正向硅胶柱层析、Sephadex LH-20凝胶柱层析、RP-18柱层析、薄层制备等方法,从Sarcodon joedes子实体甲醇提取物中分离得到9个单体化合物。经氢谱(1H-NMR)、碳谱(13C-NMR)、质谱(EI-MS, ESI-MS)等波谱技术鉴定了其中的8个化合物,它们的结构分别为:ergosta-7,22-dien-3β-ol(1),麦角甾醇过氧化物(2),3β,5α-dihydroxy-6β-methoxyergosta-7,22- diene(3),(22E,24R)-3β-hydroxy-24-methylcholesta-5,22-dien-7-one(4),苯甲酸(5),对羟基苯甲酸甲酯(6),3,4-二羟基苯甲酸(7),(2S,3R,4E,8E)-1-β-D-glucopyranosyl-3-hydroxy-2-[(R)-2’-hydroxyhexadecanoyl]amino-9-methyl-4,8-octadecadiene(9)。所有化合物均为首次从Sarcodon joedes中分离得到。

【Abstract】 Sarcodon joedes, a higher fungus belonging to the class Basidiomycete, is mainly distributed in Yunnan Province of China. In recent years, great attention has been paid to the genus Sarcodon, and many secondary metabolites with new structures and biological activities have been discovered at home and abroad. However, there is no report on the chemical constituents and biological activities of Sarcodon joedes.In this thesis, the chemical constituents from the fruiting body of Sarcodon joedes were studied, and the research progress in chemical constituents and biological activity of higer fungi was overviewed.The MeOH extract from Sarcodon joedes was subjected to repeated chromatographic columns such as silica gel, Sephadex LH-20, RP-18 column chromatography and preparative TLC to afford 9 compounds, on the basis of spectroscopic analysis including 1H-NMR, 13C-NMR, EI-MS and ESI-MS, the structures of 8 compounds were elucidated as ergosta-7,22-dien-3β-ol ( 1), ergosterol peroxide (2), 3β,5α-dihydroxy-6β-methoxyergosta-7, 22-diene (3), (22E,24R)-3β-hydroxy-24-methylcholesta-5,22-dien-7-one (4), benzoic acid (5), methyl p-hydroxybenzoate (6), 3,4-dihydroxybenzoic acid (7), (2S,3R,4E,8E)-1-β-D-gluco- pyranosyl-3-hydroxy-2-[(R)-2’-hydroxyhexadecanoyl]amino-9-methyl-4,8-octadecadiene (9), respectively. All of the compounds were isolated from Sarcodon joedes for the first time.
