藏山羊胚胎毛囊的形态发生及β-catenin的表达模式

【摘要】 本研究以胚龄(Ed)51~91藏山羊胚胎为实验材料,用Ed 53~102藏绵羊胚胎和Ed77~88牦牛胚胎作为对照,通过石蜡切片、组织芯片和免疫组化方法,研究藏山羊胚胎不同部位毛囊形态发生的差异,并定性和定量检测β-catenin在不同部位毛囊形态发生中的表达。结果如下:1.藏山羊胚胎各部位毛囊形态发生的时间、密度、大小等均有差异,这种差异与皮肤下对应的骨骼类型相关。2.β-catenin和SHH在藏山羊胚胎毛囊形态发生过程中的表达至少有两种模式:一种以间充质为主导的模式(主要见于肢端),另一种是表皮为主导的模式(主要见于躯体其它部位)。3.藏山羊胚胎尾巴背面毛囊的形态发生比腹面迅速,从尾尖至尾根背腹面毛囊的数目和发育阶段均呈递减梯度;在藏山羊尾巴初次发生的毛囊中,间充质与表皮细胞数目的比值与藏绵羊差异显著。4.β-catenin在藏山羊胚胎尾巴毛囊形态发生中的表达属于表皮—间充质均衡模式,而牦牛和藏绵羊则属于表皮为主导的模式。结果表明,β-catenin信号在藏山羊毛囊形态发生过程中的表皮—间充质相互作用中起着极为重要的作用。据推测,β-catenin在表皮—间充质的表达模式的差异可能对于毛囊类型的决定具有重要作用。

【Abstract】 Ed(embryonic day) 51~91 Tibetan goat embryos, Ed 53~102 Tibetan sheep embryos and Ed 77~88 Yak embryos were used as test and control materials respectively to investigate hair follicle morphogenesis of different parts, the paraffin embed tissue section was stained by H.E method for studying the histomorphologenesis changes and EnVision immunohistochemistry for detecting qualitatively and quantitatively expression ofβ-catenin during hair follicle morphogenesis.Results is following:1. The differences of time, density and size obviously occurred in different parts of hair follicle morphogenesis in Tibetan goat, these differences relate to the skeleton types under the skin.2. There are at least two types of expression pattern ofβ-catenin and SHH during hair follicle morphogenesis, they are mesenchymal pattern(acral hair follicles morphogenesis) and epidermal pattern(other body parts hair follicles morphogenesis).3. The back of Tibetan goat embryos tail hair follicle morphogenesis rapidly than the ventral, the number and the developmental stage of hair follicle were reduced dorsally and ventrally from pointed to root. In the primary occurrence of the Tibetan goat tail hair follicle, the ratio of mesenchymal and epithelial cells number significant difference in Tibetan sheep.4. The epidermis signaling and the mesenchyme signaling dominate during hair follicle morphogenesis in Tibetan goat embryos tail, and the epidermis signaling dominate and the mesenchyme signaling is secondary during hair follicle morphogenesis in Bos grunniens embryos and Tibetan sheep embryos.Our results indicateβ-catenin functions on the epithelial-mesenchymal reciprocal interaction during hair follicle morphogenesis in Tibetan goat. We speculate that the expression pattern differences ofβ-catenin expression may play a key role in determining hair follicle types.
