FTIR、XRF、PXRD对中药品质的研究

【摘要】 论文采用傅里叶变换红外光谱法(FTIR)、X射线荧光分析法(XRF)、粉末X射线衍射法(PXRD)三种现代分析技术以及粉末显微组织鉴别法,对航天诱变育种中药材的整体内在品质特征进行了比较、分析和研究。首先,对生长旺盛、植株健壮的航天诱变育种板蓝根和防风两种药材的性状和显微组织进行了鉴别和对比分析。结果显示,太空组的板蓝根和防风的显微组织,与地面组的板蓝根和防风有明显差异,这说明航天诱变对搭载组的两种药材种子的基因产生了一定影响。其次,通过对两组板蓝根样品(搭载组和地面组)的FTIR谱和相应的二阶导数谱进行对比分析和研究,结果显示,太空组板蓝根中多种活性成分有所增加。最后,对两组板蓝根样品的晶体物质和多种元素指标进行了测定和分析。对两组样品的XRF谱对比分析,发现太空组板蓝根与地面组板蓝根所含的元素种类并没有明显改变,但多种元素的含量却有很大差异。由两样品的PXRD谱可知,两组样品的晶体物质含量都比较少,且太空组的晶体物质含量更少。论文通过对药材样品,从粉末显微组织、分子、多种元素以及晶体物质四个层面进行分析、研究和表征,表明太空组板蓝根的多指标活性成分较地面组明显优化。

【Abstract】 In this paper, Powder Micro-image Identification and three modern parsing techniques (Fourier transformation infrared spectrometry, X-ray fluorescence and Powder X-ray diffraction) has carried on the comparison, analysis and research to spaceflight mutagenesis breeding traditional Chinese medicine’s whole intrinsic quality characteristic.First, the spaceflight mutagenesis breeding Radix Isatidis and Saposhnikovia divaricata which have advantages on appearance traits such as grow well and strong, were identified and comparative analyzed by using Powder Micro-image Identification. The results showed that the micro-structure of them were different from the ground groups (the ground Radix Isatidis and ground Saposhnikovia divaricata), which indicating the genes of Radix Isatidis and Saposhnikovia divaricata seeds have been effected by the space environment when they were carried into space.Second, the FTIR spectra and the corresponding second derivative FTIR spectra of Radix Isatidis samples (both space group and ground group) were analyzed and researched. It showed that multi-index active ingredients have increased in the space group. FTIR was suitable for analyzing the whole components of the spaceflight mutagenesis breeding traditional Chinese medicine which has large variation extent and quick stability of traits.Finally, crystal material and a variety of elements of the two groups were determined and analyzed. From the XRF spectra, it can conclude that the kinds of elements contained in the two groups were almost the same, but the percentages of those elements were obviously different. It can conclude that the content of crystal material of the two samples was both low, and the space group has less from PXRD spectra.It can conclude that the multi-index active ingredients of space Radix Isatidis have optimized by the analysis、research and characterization of the medicinal herbs from the four dimensions such as powder microstructure, molecule, multi-element and crystal material.
