三七芦头皂苷的闪式提取与分离及活性皂苷的生物转化研究

【摘要】 目的:比较并优选三七芦头总皂苷闪式提取的最佳提取以及纯化工艺;分离鉴定三七芦头中的化学成分;利用从种植人参的土壤中分离鉴定的稀有菌种Fusarium saccha-ri对三七芦头中的主要活性成分人参皂苷Rg-1进行微生物转化。材料与方法:以三七总皂苷的含量为测定指标,测定和比较不同提取工艺和纯化方法中三七总皂苷的含量;三七芦头原皂苷通过大孔树脂、硅胶柱色谱、制备液相等方法进行分离,得到化合物结构经波谱分析鉴定;转化产物通过制备液相进行分离。结果:1.用10倍量70%的乙醇,闪式提取2次所得三七芦头总皂苷的含量高于传统的乙醇回流法和超声法;经过D-101树脂纯化后的三七总皂苷含量高于其他两种方法。2.从三七芦头中分离得到13个化合物,分别鉴定为人参炔三醇、20(R)-人参皂苷-Rh1、人参皂苷-Rh2、人参皂苷-F1、人参皂苷-Rg-1、人参皂苷-Rg2、人参皂苷-Rg3、人参皂苷-Re、人参皂苷-Rd、人参皂苷-Rb1、人参皂苷-Rb3、β-谷甾醇、胡萝卜苷。3.首次从人参皂苷Rg1的Fusarium sacchari转化产物中分得3个化合物,分别鉴定为20(R)-人参皂苷-Rh1、人参皂苷-F1和20(S)-原人参三醇。结论:1.采用闪式提取是一种比较经济和有效的三七芦头总皂苷的提取工艺。2.化合物Ⅰ为首次从三七芦头中分离得到的微量成份。3.Fusarium sacchari可转化三七芦头中人参皂苷-Rg1为20(R)-人参皂苷-Rh1、人参皂苷-F1和20(S)-原人参三醇,是一种具有开发价值的稀有活性菌株。

【Abstract】 Purpose: To compare and optimize the extraction and purification techniques oftotal saponins in the Rhizome saponin of Panax notoginseng; To study the chemical constituents from the Rhizome saponin of Panax notoginseng; Active microbial strains were isolated from soils cultivated ginseng and the strain was identified as Fusarium sacchari, transformation of ginsenoside Rg1 by Fusarium sacchari from the Rhizome saponin of Panax notoginseng.Material and method: Take the content of total saponins of the basal part of stem and beard from Panax notoginseng as index,determine and compare the contents of total saponins that was determined by spectrophotometric method through the different extraction process. The compounds separated by the process of silica gel column, gelatum column and prep HPLC; Transforming products separated by the processof silica gel column,gelatum column and prep HPLC.Results:1. With adding 10 times water for smashing tissue extracting 2 min in the first timeand for 1 min in the second,the content of total saponins of the basal part of stemand beard from Panax notoginseng was higher than ethanol refluxing extraction.Thecontents of total saponins purified by D-101 macroporous absorption resin is higherthan the other two methods.2.Thirteen constitutents were identifyed as panaxytriol,ginsenoside-F1,20(R)-ginsenoside-Rh1,ginsenoside-Rh2,ginsenoside-Rg1,ginsenoside-Rg2,20(R)-ginsenoside-Rg3, ginsenoside-Re,ginsenoside-Rd,ginsenoside-Rb1,ginsenoside-Rb3,β-sitosterol and daucosterol.3.Transforming products were identifyed as ginsenoside-F1,20(R)-ginsenoside-Rh1 and 20(S)-protopanaxatriol.Conclusion:1. Smashing tissue extraction is an economic and effective methods for extraction total saponins of the basal part of stem and beard from Panax notoginseng.2. Compound I was isolated from the Rhizome saponin of Panax notoginseng for the first time.3. Transformation of ginsenosides Rg1 by Fusarium sacchari from the Rhizome saponin ofPanax notoginseng, the rarity activity microbial strains have exploitation value.
