厚朴麻黄汤对哮喘大鼠一氧化氮、内皮素1与肥大细胞脱颗粒的影响

【摘要】 目的:本实验研究从病理学、免疫组织化学和分子生物学等多角度探讨中药厚朴麻黄汤和西药氨茶碱对卵蛋白致Winster大鼠支气管哮喘模型作用机制的研究,并比较其优势。材料与方法:1.采用皮下多点注射OVA(卵蛋白)、腹腔注射百日咳杆菌菌苗的方法复制大鼠支气管哮喘模型。2.本实验按体重查随机数字表随机分为五组,正常组、模型组OVA(卵蛋白)、厚朴麻黄汤高剂量组、厚朴麻黄汤低剂量组和氨茶碱组。3.高、低剂量厚朴麻黄汤对OVA(卵蛋白)致大鼠支气管哮喘肺组织形态的研究通过观察分析模型大鼠一般状况、HE染色等方法,观察各组肺组织病理学的变化,对大鼠支气管哮喘模型进行评价,从而研究厚朴麻黄汤对支气管哮喘模型大鼠的作用机制,并比较各治疗组在整体状态和局部肺组织的不同。4.高、低剂量厚朴麻黄汤对OVA(卵蛋白)致大鼠支气管哮喘血液中NO变化的研究通过采用硝酸还原酶法,研究NO在支气管哮喘大鼠血液中的变化,以及高剂量、低剂量厚朴麻黄汤对其的影响比较,探讨厚朴麻黄汤治疗支气管哮喘的作用机制。5.高、低剂量厚朴麻黄汤对OVA(卵蛋白)致大鼠支气管哮喘血液中ET-1变化的研究通过采用放射免疫法,研究ET-1表达在支气管哮喘大鼠血液中的变化,以及高剂量、低剂量厚朴麻黄汤对其的影响比较,探讨厚朴麻黄汤治疗支气管哮喘的作用机制。6.通过显微镜镜下观察肥大细胞脱颗粒的情况,进一步探讨厚朴麻黄汤治疗支气管哮喘的作用机制。结果:1.成功复制大鼠支气管哮喘模型;2.厚朴麻黄汤能明显改善支气管哮喘大鼠的整体状态、减少支气管和肺组织中炎症浸润及气道痉挛,从组织结构上减轻支气管哮喘的程度。3.高、低剂量厚朴麻黄汤均可以降低NO和ET-1水平,减少气道炎性因子的释放,降低支气管哮喘程度。4.高、低剂量厚朴麻黄汤均降低了肥大细胞脱颗粒百分率,有效控制了大鼠变态反应的程度。结论:1.采用皮下多点注射卵蛋白(OVA)的方法,成功诱导大鼠支气管哮喘模型。2.各治疗组明显改善大鼠的一般状态、减轻肺组织的炎症细胞浸润程度,其中厚朴麻黄汤高、低剂量组的治疗效果无明显差异。3.高、低剂量厚朴麻黄汤均可降低大鼠血液中NO、ET-1水平和腹腔肥大细胞脱颗粒百分率,作用效果与氨茶碱接近。从而进一步探讨经典方剂厚朴麻黄汤治疗支气管哮喘的药效机制。

【Abstract】 Purpose:This study from the pathological morphology,immunohistochemistry and molecular biology,such as the perspective of Chinese medicine and western medicine Mahuangtang Honokiol Aminophylline on ovalbumin-induced bronchial asthma model in rats Winster mechanism research,and compare its position.Material and method:1.Using multi-point injection subcutaneous OVA(Ovalbumin),intraperitoneal injection of Bordetella pertussis vaccine approach rat model of bronchial asthma.2.In this experiment according to the weight check random number table were randomly divided into five groups,the normal group,model group OVA (ovalbumin),Magnolia Mahuangtang high-dose group,Honokiol Mahuangtang aminophylline group and low-dose group.3.High-and low-dose ephedra Magnolia Decoction on OVA(ovalbumin)-induced rat lung tissue morphology of bronchial asthma researchRat model by observing and analyzing the general situation,HE staining and other methods,to observe the group of pathological changes in morphology of the rat model of bronchial asthma evaluation,which studies Mahuangtang Honokiol rat model of bronchial asthma the role of mechanisms,and compare the treatment groups in the overall state and local organizations,different.4.High-and low-dose ephedra Magnolia Decoction on OVA(ovalbumin)-induced asthma in rats the expression of NO in blood researchThrough the use of nitrate reductase method to study the NO expression in rats with bronchial asthma changes in lung tissue,as well as the high-dose, low-dose Mahuangtang Honokiol the impact of its comparison,to further explore Mahuangtang Honokiol from immunohistochemistry on bronchial the impact of asthma. 5.High-and low-dose ephedra Magnolia Decoction on OVA(ovalbumin)-induced bronchial asthma in the blood of rats with ET-1 expression studiesThrough the use of radioimmunoassay to study the ET-1 expression in bronchial asthma in rats changes in lung tissue,as well as the high-dose, low-dose Mahuangtang Honokiol the impact of its comparison,to further explore Mahuangtang Honokiol from immunohistochemistry on the effects of bronchial asthma.Were observed through the microscope degranulation of mast cells further from the perspective of molecular biology to explore the treatment of Honokiol Mahuangtang mechanism of bronchial asthma.Results:1.The success of rat model of bronchial asthma;2.Honokiol Mahuangtang bronchial asthma can significantly improve the overall state of rats to reduce the bronchial and lung tissue inflammation and airway infiltration cramps,from organizational structure to reduce the extent of bronchial asthma,high-dose group than low-dose group;3.High-and low-dose Mahuangtang Honokiol may reduce the NO and ET-1 expression, and high-dose group for the excellent Mahuangtang Honokiol improve bronchial and lung tissue inflammation and airway spasm infiltration,reducing the degree of bronchial asthma,and inflammatory depth factor aspects of the pathogenesis of bronchial asthma;4.High-and low-dose Mahuangtang Honokiol were reduced rate of mast cell degranulation,effective control of the degree of allergic rats.Conclusion:1.Multi-point subcutaneous injection using ovalbumin(OVA) approach,the success of induced rat model of bronchial asthma.2.Treatment groups significantly improved the general state of rats to reduce the lung tissue of the degree of inflammatory cell infiltration,which Honokiol Mahuangtang high-and low-dose treatment group no significant difference.3.High-and low-dose ephedra Honokiol rat soup may lower blood NO,ET-1 levels and peritoneal mast cell degranulation percentage,with the effect of aminophylline close.Through the treatment process can reduce the airway and lung tissue infiltration of inflammatory cytokines,improve alveolitis, effective to alleviate airway edema,convulsions narrow.Inflammatory factors from the perspective of depth Mahuangtang Honokiol classic prescription treatment of bronchial asthma pathogenesis.
