超短波治疗对大鼠骨骼肌缺血再灌注损伤愈合的影响

【摘要】 目的通过检测IGF-1在大鼠骨骼肌缺血再灌注损伤模型中的表达及观察局部愈合情况了解超短波治疗对骨骼肌缺血再灌注损伤愈合的作用。方法取健康成年SD大鼠24只,体重大致相等。应用随机数字表法选取6只作为空白组(O组),余下18只采用外科手术游离股动脉后夹闭制作实验性骨骼肌缺血再灌注损伤模型。造模成功后,按前法随机分为I/R、I/R无热量、I/R微热量三组,I/R无热量组、I/R微热量组依次给予10分钟不同强度的超短波治疗,两组均治疗6天。各鼠均于8天后行颈椎脱臼法处死,切开术侧皮肤,显露腓肠肌,观察肌肉损伤愈合情况。观察完毕后取术侧腓肠肌,进行HE染色及免疫组织化学染色,观察肌肉肿胀程度、肌纤维排列情况、新生肌细胞生长情况及IGF-1的表达强度的差异。结果1、肉眼观察:O组为正常肌肉组织。I/R组肌肉组织肿胀较明显,可见散在出血点,I/R无热量、I/R微热量组肌肉组织未见明显肿胀和出血点。2、HE染色光镜下观察:O组肌纤维排列整齐,I/R、I/R无热量、I/R微热量组间比较肌纤维排列有差异。I/R组中新生肌细胞和新生血管少于I/R无热量、I/R微热量组。3、IGF-1表达:I/R无热量、I/R微热量组标本中IGF-1表达较O、I/R组高,有统计学意义(P<0.05),I/R微热量组较I/R无热量组的IGF-1表达更高,有统计学意义(P<0.05)。结论1、超短波治疗可能通过增加IGF-1表达而促进骨骼肌I/R损伤愈合。2、超短波治疗骨骼肌I/R损伤有效,且微热量治疗比无热量治疗效果更佳。

【Abstract】 ObjectivesThrough the detection of IGF-1 in rat skeletal muscle ischemia-reperfusion injury model, expression and observation to understand the ultrashort wave therapy on skeletal muscle ischemia-reperfusion injury in the healing role.MethodsCheck the health of 24 adult SD rats,weighing roughly equal. Using random number table to select 6 as the blank group (O group), 18 the use of the remaining free surgery after femoral artery occlusion producing experimental skeletal muscle ischemia-reperfusion injury model. After the success of the model, according to the former method were randomly divided into I/R, I/R without heat, I/R Micro heat three groups,I/R group without heat, I/R group of micro-heat followed by 10 minutes to give varying degrees of ultrashort wave treatment.The two groups were treated six days.The mice were 8 days after cervical dislocation line method executed operative side skin incision,exposed gastrocnemius observed muscle injury healing. Observation of the operative side after the gastrocnemius check for HE staining and immunohistochemical staining to observe the degree of muscle tumefaction ,muscle fiber with the situation,myogenic cells in the growth of new and the expression of IGF-1 intensity differences.Results1. macroscopic observation: O group was as normal muscle tissue. I/R group with more pronounced tumefaction of muscle tissue can be seen scattered bleeding points, I/R without heat, I/R group of micro-heat muscle tissue tumefaction less and no obvious bleeding point.2.HE staining were observed: O group of muscle fibers were arranged regularly.I/R,I/R without heat,I/R micro-heat group compared with the difference between muscle fibers and I/R group had less myogenic cells in the growth of new and neovascularization than I/R without heat, I/R micro-heat group.3. The integrated optical density(IOD) of IGF-1: I/R withoutheat, I/R group of micro-heat samples IGF-1 expression than the O, I/R group,there is statistical significance (p<0.05). I/R group of micro-heat than in I/R without heat group IGF-1 expression more , there is statistical significance (p<0.05).Conclusions1.Ultrashort wave treatment may increase IGF-1 expression and thepromotion of skeletal muscle I/R injury to heal.2.Ultrashort wave Treatment of skeletal muscle I/R injury in an effective and micro-heat treatment than that without heat treatment better.
