卡介苗菌DNA抗肿瘤免疫的实验研究

【摘要】 目的:通过检测三组荷瘤裸鼠血清IFN-γ和TNF-α水平,NK、Mφ细胞杀伤活性,测定卡介苗菌DNA疫苗体内对肿瘤抑制率和对不同肿瘤细胞的体外抑制率,比较卡介苗菌制剂DNA与卡介苗的抗肿瘤免疫效果并探讨其免疫机制。方法:从卡介苗中纯化提取卡介苗菌DNA疫苗,采用细胞培养移植法,复制荷瘤裸鼠模型,将24只荷瘤裸鼠随机分为TES(2-三(羟甲基)甲基-3-氨基丙烷磺酸)组、卡介苗(BCG)组、DNA组,每组8只。于接种A549细胞株后第15天,TES组、BCG组、DNA组分别腹腔注射TES液、BCG液、DNA液各0.3 mL,隔2 d 1次,共8次。采用双抗体夹心酶联免疫检测法检测3组荷瘤裸鼠血清IFN-γ、TNF-α水平,及NK、Mφ细胞杀伤活性;卡介苗菌DNA疫苗体内对抑制率、对不同肿瘤细胞体外抑制率的测定。结果:卡介苗菌DNA制剂、BCG可抑制肿瘤生长。卡介苗菌制剂DNA可增强荷瘤裸鼠NK和Mφ杀伤活性,且其差异具有统计学意义P<0.05;及调节血清IFN-γ、TNF-α水平,且其差异具有统计学意义P<0.05。结论:卡介苗菌制剂DNA与卡介苗均有抗肿瘤免疫特性,而且卡介苗菌DNA制剂抗肿瘤免疫作用强于卡介苗。

【Abstract】 Objective: To examine nude mice serum levels of IFN-γandTNF-αof.3 groups, to detect killing activity of NKand Mφcell and the suppression rate of tumor in vivo and different tumor cells in vitro , To compare Bacillus Calmette Guerin DNA with Bacillus Calmette Guerin on antitumor of effects and to explore the mechanism of immunityMethods: Bacillus Calmette-Cuerin DNA was extracted and purified from the BCG vaccine , the tumorbearing models of nude mice were established, 24 tumor bearing nude mice were randomly divided into TES,BCG and DNA groups and each group had 8 mice. 15 days after Vaccination A549,The mice in TES group,BCG group and DNA group were injected peritoneally with TES,BCG and DNA solution.Each tumor bearing nude mice was injected 0.3 mL,once every 2 days,a total of 8 times.The serum levels of IFN-γ, and TNF-α. were detected by enzyme linked immunosorbent assay,and killing activity of NK, Mφcell of three groups tumor bearing nude mice,the suppression rate of tumor in vivo and different tumor cells in vivo were detected.Results:BCG and DNA can suppress the tumor growth,NKMφcell killing activity were enhanced by DNA,and the difference were statistically significant , P <0.05;Bacillus Calmette-Cuerin DNA may adjust nude mice serum levels of IFN-γ,TNF-α., and the difference were statistically significantP<0.05.Conclusions: DNA and the BCG vaccine all have the characteristics of antitumor immune, but antitumor immunity effect of Bacillus Calmette-Cuerin DNA is stronger than of BCG..
