奶牛酮病检测试纸条的研制与应用

【摘要】 本文通过对奶牛酮病检测方法的研究,依据硝普盐法检测酮体原理,开发研制出应用于尿酮体检测的浸渍试纸,并在此基础上研制出应用于乳酮体检测的改良试条,旨在提高对奶牛酮病早期诊断的有效监测,降低治疗成本。通过对所研制成品的初步应用研究,明确了应用浸渍试纸与改良试条结合测定酮体的方案,可用于亚临床酮病早期监测,为今后推广和应用该产品提供科学依据。本试验设计了完整的浸渍试纸的制备方案,合理设置制备工艺,科学制作了标准显色卡,验证实验检测结果以自制浸渍试纸显色深浅由深到浅排序,与测定的酮体含量正相关,与酮粉法、美国URS-1K酮体检测试纸对照总符合率达93.94%和96.97%;但是本实验研制的浸渍试纸对尿酮敏感,对乳酮不敏感,通过实验研究证实了酮病牛尿酮含量大于乳酮含量,乳酮含量是检测酮病牛酮体含量的重要参数,为更有效的应用于酮病的临床监测奠定基础。基于浸渍试纸设计理论设计了改良试条的设计方案,采取先制作塑料模型,再定量添装显色粉剂,实现组装试条来检测乳酮,明确了制备工艺。本实验研制的改良试条检测结果显色深浅由深到浅排序,与乳汁中酮体含量正相关,改良试条与酮粉法对照总符合率达92.93%,且酮体含量越高符合率越高。说明所研制试纸和改良试条可有效检测临床酮病,对隐性酮病有较好的半定量检测效果。最后通过两种试纸的临床应用研究,选取实验牛产后第3d、7d、15d、45d四个观测点进行验证,实验表明自制试纸和改良试条灵敏性和特异性均较好,初步确立了应用所研制试纸半定量检测尿酮、乳酮来监测酮病的方法。综合实验研究表明:所研制试纸条监测酮病具有方便快捷、易于大面积推广使用等特点,故本研究为检测奶牛尿及乳汁中酮体含量提供有效的产品和科学的方案,可以有效监测奶牛酮病的发病,提高预防效果,降低治疗成本;但是本试纸只限于半定量测定,精确度仍有待进一步的研究探讨。

【Abstract】 To improve the effective implementation of early diagnosis for cow ketosis and reduce monitoring cost, impregnated test strip of detecting urine ketone was developed by studying the testing method of cow ketosis and according to the theory of nitroprusside determining ketone, and modified test strip of detecting milk ketone was subsequently researched and developed on this basis. In this study, the method of testing ketone was established by using impregnated test strip and modified test strip, which can apply to early monitoring of subclinical ketosis and provide scientific basis for spread and application of this product.In this experiment, the preparation plan of impregnated test strip was completely designed and the preparation process was established reasonably and the standard color card was manufactured scientifically. The result of verification experiment was shown that the appearing color order of self-made test strip was from dark to light, which presented positive correlation as the testing content of ketone and the total coincidence of 93.94% and 96.97% compared with ketone body powder method and American URS-1K test strip respectively. However, the impregnated test strip was sensitive to urine ketone, insensitive to milk ketone. The urine ketone content of cow ketosis was more than that of milk ketone, and the content of milk ketone was an important parameter of representing ketone body content of cow ketosis, which can be used to monitoring the clinical ketosis effectively. Basing on the theory of impregnated test strip, the scheme of modified test strip was designed that the plastic model was manufactured firstly and then colored power was installed, which realized assembled test strip to detect milk ketone and identified the preparation process. The detecting result of modified test strip showed that the appearing color order was from dark to light and presented the positive correlation with milk ketone content. The coincidence reached 92.93% compared with modified test strip and ketone body powder method in the detecting milk ketone, and the higher content of ketone was, the higher coincidence of color was, which indicated the developing two test strips can effectively detect clinical ketosis and had the better semi-quantitative effective to subclinical ketosis.To observe the clinical application of two kinds of test strip, cows affected ketosis were chosen at postpartum 3d, 7d, 15d, and 45d. The results showed that the sensitivity and specificity of self-made test strips were both better and preliminarily established the methods of semi-quantitative detecting urine ketone and milk ketone of cow ketosis.In a word, self-made test strips of cows ketosis had characteristic of convenience, rapid diagnosis, and easy application, etc. So, this study will provide effective product and scientific scheme for detecting the content of urine ketone and milk ketone, meanwhile it can monitor the pathogenic process of the cow ketosis effectively, enhance prevention effect and reduce the treatment cost. But the strips only restrict in the semi-quantitative determination, and still needs more research in the accuracy in future.
