植物生长调节剂对大豆根系建成的调控效应

【摘要】 试验于2008年在黑龙江省大庆市八一农垦大学试验基地进行,以K4(垦农4号大豆)和H25(合丰25号大豆)为实验材料,幼苗采用矿栽砂培方式培养,通过植物生长调节剂DTA-6和S3307在不同浓度条件下对两品种大豆的种子进行浸种处理,研究根系建成期大豆根系形态结构变化,探讨植物生长调节剂对根系生理生化的调控机制及解剖、超微结构的影响,筛选最佳浓度。主要研究结果如下:1适宜浓度(25mg/L-50mg/L)条件下的DTA-6和S3307(0.1mg/L-0.4mg/L)对根建成期的大豆根系形态均不同程度的起到了正向调控作用。其中,D50(50mg/L的DTA-6)和S0.4(0.4mg/L的S3307)的处理效果最佳。D50处理对K4和H25品种的主根发育起到了正向调控作用。明显促进了K4的侧根长度,增加了侧根数,根体积和根干重。S0.4处理对K4的侧根长度,根体积,根干重的调节效果较好,对H25的根干重的调节效果达最佳。而DTA-6在100mg/L(D100)浓度条件下抑制了主根和侧根的生长发育。2两品种经适宜浓度的DTA-6和S3307浸种后,显著的提高了根系活力,增加了根系中的可溶性蛋白,硝态氮和可溶性糖的含量。其中,D50和S0.4处理效果较好,均明显提高了K4根系和H25大豆根系可溶性蛋白含量,硝态氮含量和可溶性糖含量,且D50处理对K4根系活力的提高起正向调控作用。D100处理减少了大豆根系的可溶性蛋白,硝态氮和可溶性糖含量。3 D25和S0.4处理更有利于根系酶活性的提高和MDA含量的降低,且效果达最佳。其中,D25处理显著的提高了K4根系INV活性、POD活性、SOD活性,降低了MDA的含量,对H25根系POD活性的提高,MDA含量的降低均起到了正向调控作用。而S0.4处理则提高了K4根系INV活性,降低了MDA含量,对H25根系INV活性、MDA含量的降低调控效果较好。4 D50和S0.4处理效果较好。D50明显增加了K4大豆根系直径长度,根系导管数目,明显扩大了根系输水截面积和韧皮部截面积,增加了H25的根系导管数目,根系输水截面积和根系韧皮部截面积。S0.4处理显著增加了K4根系直径长度,中柱鞘直径长度,输水截面积以及根系韧皮部截面积。对H25根系中柱鞘直径长度和韧皮部截面积的增加效果最佳。5经D50和S0.4处理后的液泡、线粒体及淀粉质体等结构明显而且清晰,线粒体、淀粉质体、质体数量丰富(图版7-1和7-2)。且均在不同时期出现高尔基体,并有少量内质网出现。D100和S0.2对大豆根细胞发育的抑制作用较明显。

【Abstract】 The experiments were conducted in Daqing city, Hei Long jiang Province, in 2008. Kennong 4 and Hefeng 25 was selected as the experimental material, seedling was cultured by sand, soaking se- eds with two different PGRs(DTA-6 and S3307)in different thickness of soybean. Studied the changes of morphology and structure in the period of root building in soybean, researched the effect of PGRs on the physiology and biochemistry, anatomical structure, ultrastructure, selected the best thickness. the main results were:1 Appropriate thickness of DTA-6(25mg/L ~ 50mg/L)and S3307 regulated the morphology of Soybean root forward. The treatments of D50 and S0.4 are the best. D50 regulated the main root length of K4 and H25 forward. Promoted the length and the number of the lateral root, volume and dry weight of root previously. Effects of S0.4 on the length of the lateral root, volume and dry weight of root are better, and on dry weight of root of H25 are the best. But D100 inhibited the main root and the lateral root length.2 After the two varieties soaking seeds with appropriate thickness of DTA-6 and S3307, increased the root activity, the content of the soluble protein, nitrate nitrogen and soluble sugar. D50 and S3307 are the best, increased the the content of the soluble protein, nitrate nitrogen and soluble sugar of K4 and H25 previously, and D50 regulated the root activity forward. D100 reduced the content of the soluble protein, nitrate nitrogen and soluble sugar.3 D25 and S0.4 can increased the activity of enzyme and reduce the content of MDA effectively, and are the best. D25 increased the activity of INV, POD, SOD, reduced the content of MDA of K4, regulated the activity of POD and the content of MDA of K4 forward. S0.4 increased the activity of INV, reduced the content of MDA of K4, regulated the activity of INV, the content of MDA.4 D50 increased the length of the diameter, the number of catheter in soybean root, the sectional area of root xylem and phloem of K4 previously, and increased the the number of catheter, the sectio- nal area of root xylem and phloem of H25. S0.4 increased the length of the diameter and the pericycle, the sectional area of root xylem and phloem of K4 previously, effectively increased the length of the pericycle and sectional area of phloem.5 After treated by PGRs of D50 and S0.4,the structure of vacuole, mitochondria and starch grain, etc are distinct, the number of mitochondria, starch grain and plastid are also rich in it(chart 7-1 and 7-2). Golgi apparatus and a spot of endoplasmic reticulum appeared in different period. D100 and S0.2 inhibited the development of cell effectively.
