新型噻唑烷酮和酰基硫脲衍生物的合成

【摘要】 噻唑烷酮类化合物具有重要生理活性,可作杀菌剂、放射线增感剂、抗高血压和糖尿病的药物。酰基硫脲、1,2,3-三唑以及1,3,4-噻二唑类衍生物具有舒张血管、抗癌、抗病毒、杀菌以及抗痉挛等特殊的生理活性,成为目前硫脲和唑类化合物研究的一个新热点。研究表明,若将1,2,3-三唑基引入到某些活性分子中,如青霉素、头孢菌素等,能显著改善母体分子的生物活性。鉴于具有药效基团的杂环化合物进行活性叠加后,可明显增强其药物活性的特点。我们将1,2,3-三唑基和二苯羟乙酰基引入到酰基硫脲和噻唑烷酮衍生物中,通过超声辐射法促进反应,合成出一系列未见文献报道的新型噻唑烷酮和酰基硫脲衍生物,为药物筛选提供先导化合物。超声辐射法因具有操作方法简单,反应条件温和,缩短反应时间,提高反应产率等特点,所以近二十年来在有机合成中的应用发展非常迅速。论文主要分为以下两部分:第一文献综述部分对噻唑烷酮衍生物、硫脲衍生物、1,2,3-三唑、1,3,4-噻二唑及二苯羟乙酸类化合物的研究进展进行了较为系统地阐述,简要介绍了超声辐射的原理及其在有机合成中的应用及Knoevenagel反应研究的新进展。第二实验部分1.采用超声辐射法合成4-(2-苯基-2H-1,2,3-三唑-4-基)-5,6-二氢-5-氧-6-芳亚甲基-噻唑并[2,3-c][1,2,4]三唑;2.采用超声辐射法合成N-芳酰基-N’-[5-(2-苯基-1,2,3-三唑-4-基)-1,3,4-噻二唑-2-基]硫脲;3.采用超声辐射法合成N-芳酰基-N’-二苯羟乙酰氨基硫脲的合成。

【Abstract】 Thiazolidone derivatives also have very important physiological activities, and they can be used as a fungicide, radiation sensitizing agents, antihypertensive drugs and diabetes.Acylthiourea, 1,2,3-triazole, and 1,3,4-thiadiazole derivatives have special physiological activities, such as relaxing blood vessels, anticancer, antivirus and antispasm. It has become a new hot spot in studies of azole compounds. The studies in recent years have shown that introducting the 1,2,3-triazole-based to some active molecules can significantly improve the biological activity of matrix elements.According to the principle of superposition activity, in this article, we have synthesized a series of thiazolidone and acylthiourea derivatives which are not reported in the literature by introducting 1,2,3-triazole-based, 1,3,4–thiadiazole and dibenzo into acetyl-hydroxy acyl thiazolidoneand thiourea derivatives through the promotion of ultrasonic radiation. This can provide leading compounds for new drug screening.Because of simple operation, mild reaction conditions, short reaction time, higher yield, the development of Ultrasonic radiation in the organic synthesis research has been very fast in recent years.This thesis consists of two major parts:The first part is about literature review. The principle and application of ultrasonic irradiation in organic synthesis were simply introduced. Furthermore, it symtematically expounded the research progress of thiazolidone derivatives, thiourea derivatives, 1,2,3-triazole, 1,3,4-thiadiazole and benzilic acid compounds.The second part is experimental content.1.4-(2-phenyl-1,2,3-triazol-4-yl)-5,6-dihydro-5-oxo-6-arymethylidene-thiazol-[2,3-c][1,2,4]triazole were synthesized by Ultrasonic radiation.2. N-aroyl-N′-[5-(2-phenyl-1,2,3-triazol-4-yl)-1,3,4-thiodiazol-2-yl]thioureas derivatives were synthesized by Ultrasonic radiation.3. N-aroyl-N′-(2-hydroxy-2,2-diphenylacetamido) thioureas derivatives were synthesized by Ultrasonic radiation
