活化小胶质细胞移植后脊髓损伤大鼠神经功能恢复及评价

【摘要】 目的:研究活化小胶质细胞移植是否对大鼠受损伤脊髓有修复作用,活化小胶质细胞移植后对大鼠进行神经功能评价,以观察治疗效果。并对常用的BBB评分、改良Tarlov评分、斜板试验和运动诱发电位检测进行比较,寻求准确可靠的评价方法。方法:采用自制改良Allen脊髓打击装置制作脊髓损伤大鼠动物模型,分为对照组和移植组。移植组术后1周进行活化小胶质细胞移植,而对照组不移植。移植术后第1、2、3、4、6、10周分别采用BBB评分、改良Tarlov评分、斜板试验和运动诱发电位检测对大鼠神经功能进行评价,并进行统计学分析。结果:斜板试验测定角度在第1-6周时,均有所减小(P<0.05),但第10周未见变化(P>0.05):改良Tarlov评分第1、2、3、4周时,分值间的差别有统计学意义(P<0.05),第6、10周时,差别没有统计学意义(P>0.05);而BBB评分和MEP检测术后各周的差别均具有统计学意义(P<0.05)。结论:活化小胶质细胞移植可促进脊髓损伤大鼠神经功能的恢复;BBB评分和MEP检测对大鼠脊髓损伤后神经功能评价更具有优势。

【Abstract】 Objective:to study whether there is repair function to spinal cord injury in rats after activated microglia transplantation,to evaluate the neurological function in rats after activated microglia transplantation in order to observe the treatment results.And compare BBB scale,modified Tarlov grading, inclined plane method and motor evoked potentials test(MEPT) commonly used,to find accurate and reliable evaluation methods.Methods:We established the spinal cord injury rat models with the selfmade reformed Allen’s strike equipment,randomly divided into two groups: the transplantation group and the control group.We transplanted activated microglia to the transplantation groupbut but did not transplant to the control group.The models were scored the nerve function by BBB scale in weeks 1, 2,3,4,6and10 after the transplantation operation respectively,at the same time,inclined plane method,Modified tarlov grading and motion evoked potential detection was also carried out,and statistics analysis was conducted.Results:Critical angle in incline plane test was decreased in 6 weeks(P<0. 05),but in 10w this change disappeared(P>0.05);Modified Tarlov grading was compared:1w,2w,3w,4w,the difference of scale was different(P<0. 01),but in 6w and 10w this change disappeared(P>0.05);In every week after operation,statistical significance of the distinction of BBB scale and motion evoked potential detection existed(P<0.05).Conclusion:activated microglia transplantation can promote the recovery of the nerve function in SCI rats;for these different neurological evaluation methods,BBB scale and motion evoked potential detection have more advantages.
