不同低氧训练方式对大鼠下丘脑β-EP及血液白细胞计数的影响

【摘要】 目的:本实验旨在研究不同低氧训练方式下大鼠下丘脑弓状核β-内啡肽基因表达及下丘脑β-EP蛋白含量的变化情况,以及与血液白细胞总数及其分类百分比变化的关系。为明确β-EP对机体免疫功能调节作用的机理提供参考。研究方法:健康雄性SD大鼠75只随机分为五组,按照实验设计,进行四周不同海拔的训练,于末次训练后次日晨取血取材,进行下丘脑弓状核β-EP基因表达、下丘脑β-EP蛋白质含量的测定以及血液白细胞总数和分类百分比的测定。结果:1.低氧环境下,与HiCo组相比,HiHi组、HiLo组的大鼠下丘脑弓状核β-EP基因反应阳性颗粒OD及IOD值均增加(P<0.05及P<0.01),与HiLo组相比,HiHi组的OD及IOD值均增加明显(P<0.05及P<0.01)。2.常氧环境下,LoLo组与LoCo组相比,下丘脑β-EP蛋白含量增加(P<0.01);低氧环境下,与HiCo组相比,HiHi组、HiLo组下丘脑β-EP蛋白含量均增加(P<0.05及P<0.01),HiHi组与HiLo组相比,下丘脑β-EP蛋白含量增加明显(P<0.01);HiCo组与LoCo组相比,下丘脑β-EP蛋白含量显著增加(P<0.01),LoLo组与HiLo组相比,下丘脑β-EP蛋白含量则显著下降(P<0.05)。3.就白细胞总数而言,HiCo组与LoCo组相比,白细胞总数显著下降(P<0.01),LoLo组与HiLo组相比,则白细胞总数明显增加(P<0.01)。其分类计数百分比未发现显著性变化。4.常氧和低氧环境下,下丘脑β-EP蛋白含量与白细胞总数及分类计数百分比之间均无明显相关性。结论:1.四周实验后,五组大鼠下丘脑弓状核β-EP基因表达及下丘脑β-EP蛋白含量的变化表现为HiHi组>HiLo组>HiCo组>LoLo组>LoCo组,但下丘脑β-EP蛋白含量的变化要比下丘脑弓状核β-EP基因表达的变化更为明显。2.四周实验后,大鼠血液白细胞总数的变化表现为HiCo组明显低于LoCo组,HiLo组明显低于LoLo组;五组大鼠的白细胞分类计数百分数并未出现显著性变化。3.四周实验后,不同低氧训练方式下下丘脑β-EP蛋白含量与白细胞总数及其分类计数百分数比之间的变化并无显著相关性,β-EP对免疫功能的具体调节机制还有待于进一步研究。

【Abstract】 Objective:The purpose of the present study is to investigate the effect of the hypoxic training in different modes on the expression ofβ-ehdorphin gene in ARC and the protein concentration ofβ-EP in rats’ hypothalamus,and find out the relationship with the leucocyte counts and the percents of lymphocyte,granulocyte,monocyte.Try to illustratethe influence ofβ-EP on immune function.Method:75 8-week-old male Sprague-Dawley rats,were randomly divided into the following groups:1.living high control group;2.living high and training high group;3.living high and training low group;4.living low control group;5.living low and training low group.The rats were trained with swimming on different altitude in exercise groups basing on the experiment plan.In the morning after the last training,the rats would be Killed,and the expression ofβ-EP gene in ARC,the protein concentration ofβ-EP in rat’ s hypothalamus and the sum of leucocyte and the percents of lymphocyte,granulocyte,monocyte in blood were measured.Result:1.Compared with HiCo group,the OD and IOD ofβ-EP gene in ARC were obviously increased in HiHi group and HiLo group in anoxic condition (P<0.05 and P<0.01),and implying the expressing of mRNA in ARC was enhanced.Compared with HiLo group,the OD and IOD of HiHi group were increased(P<0.05 and P<0.01).There was no difference between other groups.2.Compared with LoCo group,the protein concentration ofβ-EP in hypothalamus were distinctly increased(P<0.01) in LoLo group;in anoxic condition,it was enhanced in HiHi group and HiLo group(P<0.05 and P<0.01),and compared with HiLo group,the protein concentration ofβ-EP in hypothalamus was increased(P<0.01) in HiHi group;compared with LoCo group,it was added(P<0.01) in HiCo group,and compared with HiLo group,the protein concentration ofβ-EP in hypothalamus was decreased(P<0.05) in LoLo group.3.Compared with LoCo group,the HiCo group’ s sum of leucocyte was obviously decreased(P<0.01),and compared with HiLo group,the LoLo group’ s leucocyte counts was evidently increased(P<0.01).There were no statistical changes between other groups. there was no significant changes in the percent of lymphocyte, granulocyte,monocyte in blood.4.in this study,between the protein concentrain ofβ-EP in hypothalamus and the percent of lymphocyte, granulocyte,monocyte had no evidently relationship in different mode training.Conclusion:1.Four weeks experiment later,the changing of the gene expression and the protein concentration ofβ-EP in five group rats behaved HiHi group >HiLo group>HiCo group>LoLo group>LoCo group, but the changing of the protein concentration was more obviously than the changing of gene expression.2.Four weeks experiment later,the leucocyte counts of HiCo group was much lower than LoCo group in rats blood,and that of HiLo group was much lower than LoLo group,there was no significant change in different count of leucocyte.3.Four weeks experiment later,there was no remarkable relationship between the protein concentration ofβ-EP in hypothalamus and the leucocyte count, the modulation mechanism betweenβ-EP and immunity could be knowm by further research in the future.
