【摘要】 目的:本课题对新疆胀果甘草抗血栓有效成分进行研究,依照体外抗血栓细胞实验结果,分离并纯化有效部位,从而得到活性单体成分并对活性成分进行含量测定,对抗血栓中药、天然药物五类新药的开发和利用提供基础资料。方法:以新疆胀果甘草水提取浸膏、60%乙醇提取浸膏、95%乙醇提取浸膏以及水提浸膏通过大孔树脂分离的流分等作为受试样品进行抗血栓活性实验,筛选有效部位;对部分有效部位进行分离;采用HPLC法对该有效部位进行含量测定。结果:1)所得到的提取物及分流中60%乙醇提取物和水提取物通过大孔吸附树脂30%乙醇洗脱部分对血栓细胞有抑制作用。2)对部分提取物进行分离,共得到8个化合物,其中甘草酸和甘草苷均对血栓细胞有抑制作用。3)采用HPLC法对不同提取物中的甘草酸,甘草次酸和甘草苷含量进行测定,其结果为水提浸膏经过大孔吸附树脂分离用30%乙醇洗脱流分中这三种成分总含量最高,其达到62.33%。结论:新疆胀果甘草对血栓细胞的增殖有抑制作用;通过活性成分的含量测定以及不同提取物和流分的体外活性筛选得到水提取物通过大孔吸附树脂30%乙醇洗脱部分为抗血栓有效部位。更多还原

【Abstract】 Objective: To research an antithrombotic active fractions of Glycyrrhiza inflata, We combined in-vitro pharmaco-dynamics experiments selected extract fraction with antithrombotic activity from these herb. According to the trails of pharmacodynamics experiments of in-vitro, we find active fractions with antithrombotic activity, so we are aimed at separating this fraction. Provide the basic information for the future development of the antithrombotic new drugs. Methods: Extracts of Glycyrrhiza inflata as samples of antithrombotic activity in vitro experiments, select and separate effective parts of the Glycyrrhiza inflata; HPLC method were used for identification and determination of the active fraction. Results: 1)There are 60% ethanol extract and 30% ethanol elution fraction inhibit the thrombosis cells. 2) On separation of effective parts, a total of 8 compounds were obtained, in which glycyrrhizic acid and glycyrrhizin have the inhibition activity of thrombosis cells. 3) Using HPLC for determine contents of glycyrrhizic acid, glycyrrhetinic acid and glycyrrhizin in extracts, contents of the 30% ethanol elution fraction is highest than other extacts, reach to 62.33%. Conclusion: 60% ethanol extract and 30% ethanol elution fraction inhibit the thrombosis cells, Through the determination of active compounds and experiments of in-vitro pharmaco-dynamics filter the antithrombotic active fractions is 30% ethanol elution fraction.更多还原
