GSPE对运动大鼠心肌自由基代谢及运动能力影响的研究

【摘要】 1研究目的通过观察运动训练和服用葡萄籽原花青素(GSPE)对大鼠心肌自由基代谢和运动能力的影响,探讨GSPE对提高机体抗氧化能力和运动能力的作用,为更好的指导运动训练、增强运动能力提供科学依据,为抗氧化新药的开发提供科学依据。2实验方法选取8周龄wister大鼠120只(实测96只),随机分为四组:A组(对照组)、B组(服药组)、C组(运动训练组)、D组(运动服药组)。C、D组大鼠经8周游泳训练,B、D组每天灌服葡萄籽原花青素。8周后,将每组按照处死的状态不同随机分为3个亚组,即:安静组、力竭即刻组、力竭恢复24小时组。并于相应时刻断头处死后取心脏测定MDA含量、SOD活性、GSH-PX活性、XOD活性、GSH含量及总抗氧化能力。测得数据用SPSS10.0统计软件进行分析,P<0.05,差异显著;P<0.01,差异非常显著。3实验结果3.1安静状态下,大鼠心肌MDA含量B组低于A组(P<0.05),D组显著低于C组(P<0.01)。SOD活性D组显著高于A、B组(P<0.01),D组高于C组(P<0.05),其余各组无显著性差异(P>0.05)。GSH-PX活性B、C组高于A组(P<0.05),D组显著高于A组(P<0.01),D组高于B、C组(P<0.05)。GSH含量B、C、D组显著高于A组(P<0.01),D组高于C组(P<0.05)。D组显著高于B组(P<0.01)。XOD活性D组稍低于其他三组但无统计学差异(P>0.05),A、B、C组无显著性差异(P>0.05)。总抗氧化能力B组高于A组(P<0.05),C、D组显著高于A组(P<0.01),D组显著高于B组(P<0.01),D组高于C组(P<0.05)。3.2力竭即刻状态下,大鼠心肌MDA含量B、C、D组显著低于A组(P<0.01),D组显著低于C组(P<0.01)。SOD活性B组高于A组(P<0.05),D组显著高于A、B、C组(P<0.01)。GSH-PX活性D组显著高于A、C组(P<0.01),B、C组高于A组(P<0.05)。GSH含量B、C、D组显著高于A组(P<0.01),D组显著高于C组(P<0.01)D组高于B组(P<0.05)。XOD活性B、D组低于A组(P<0.05),A、C组无显著性差异(P>0.05)。总抗氧化能力B、D组显著高于A组(P<0.01),D组显著高于C组(P<0.01)。3.3力竭恢复24小时状态下,大鼠心肌MDA含量B、D组低于A组(P<0.05),D组低于C组(P<0.05)。SOD活性B、D组显著高于A组(P<0.01),D组显著高于C组(P<0.01),D组高于B组(P<0.05)。GSH-PX活性B组高于A组(P<0.05),D组显著高于A、C组(P<0.01)。GSH含量B、C、D组显著高于A组(P<0.01),D组显著高于B组(P<0.01),D组高于C组(P<0.05)。XOD活性C、D组低于A组(P<0.05),A、B组无显著性差异(P>0.05)。总抗氧化能力D组显著高于A、C组(P<0.01),A、B、C组无显著性差异(P>0.05)。3.4大鼠力竭时间,D组大鼠力竭时间相比A、B两组显著延长(P<0.01);B组对比A组延长(P<0.05),D组对比C组延长(P<0.05)。4结论4.1 8周的有氧耐力训练提高了各个状态下大鼠心肌组织抗氧化能力,增强酶促系统对心肌细胞的保护作用,对心肌组织细胞起到良好的保护。4.2原花青素在主动清除自由基减少脂质过氧化反应方面表现出较大优势,还可以提高大鼠心肌组织抗氧化能力,有利于对心肌的保护。4.3有氧耐力训练联合原花青素的使用可以有效地清除大鼠心肌组织内产生的自由基,抑制脂质过氧化反应,延长大鼠运动时间,提高运动能力。

【Abstract】 1 Objective:The key point of this paper is to observing Training exercise and taking grape seed proanthocyanidin extract(GSPE) on myocardial free radical metabolism and exercise capacity, investigate the effect by GSPE on inprove the antioxidant capacity and exercise capacity,in order to give a better guide to the training exercise,and provide a scientific basis for enhance the capacity of sports and explore a antioxidant drugs.2 Method:Select 120 wister mice(meas.96),8-week-old.randomly divided into four groups:A group (control group),B group(medication group),C group(exercise training group),D group (medication group exercise).C,D group rats give eight weeks of swimming training,B,D group give GSPE every day.Eight weeks later,all mice were randomly divided into three different subgroups in accordance with the different executed status,namely:quiet group,instantly exhaustive group,exhaustive 24-hour Recovery Group.And the corresponding execution time broken heart.after the determination of MDA content in check,SOD activity,GSH-PX activity, XOD activity,GSH content and total antioxidant capacity.Measured data SPSS10.0 statistical software to conduct an independent samples T-test,compared with the control group were significantly different,P<0.05.a very significant difference,P<0.01.3 Result:3.1 In Quiet state,myocardial MDA content lower than the A group B group(P<0.05),D group was significantly lower than C group(P<0.01).SOD activity in group D was significantly higher than A,B group(P<0.01),D group was higher than C group(P<0.05),the other groups no significant difference(P>0.05).GSH-PX activity in B,C group was higher than A group(P<0.05),D group was significantly higher than the A group(P<0.01),D group was higher than B, C group(P<0.05).GSH content of B,C,D group was significantly higher than the A group(P<0.01),D group was higher than C group(P<0.05).D group was significantly higher than the B group(P<0.01).XOD activity of D slightly lower than the other three groups but no significant difference between groups(P>0.05),A,B,C group no significant difference(P>0.05).Total antioxidant capacity higher than the A group B group(P<0.05),C,D group was significantly higher than the A group(P<0.01),D group was significantly higher than the B group(P<0.01), D group was higher than C group(P<0.05).3.2 At the immediate ends of exhaustive exereise,the MDA content of myocardial B,C,D group was significantly lower than A group(P<0.01),D group was significantly lower than C group(P<0.01).SOD activity in group B than A group(P<0.05),D group was significantly higher than the A,B,C group(P<0.01).GSH-PX activity in group D was significantly higher than A,C group(P<0.01),B,C group was higher than A group(P<0.05).GSH content of B,C, D group was significantly higher than the A group(P<0.01),D group was significantly higher than the C group(P<0.01) D group was higher than B group(P<0.05).XOD activity of B,D group than in A group(P<0.05),A,C group no significant difference(P>0.05).Total antioxidant capacity B,D group was significantly higher than the A group(P<0.01),D group was significantly higher than the C group(P<0.01).3.3 At the 24 hours ends of exhaustive exercise,the MDA content of myocardial B,D group than in A group(P<0.05),D group was lower than C group(P<0.05).SOD activity in B,D group was significantly higher than the A group(P<0.01),D group was significantly higher than the C group(P<0.01),D group was higher than B group(P<0.05).GSH-PX activity in group B than A group(P<0.05),D group was significantly higher than the A,C group(P<0.01).GSH content of B,C,D group was significantly higher than the A group(P<0.01),D group was significantly higher than the B group(P<0.01),D group was higher than C group(P<0.05). XOD activity of C,D group than in A group(P<0.05),A,B group was no significant difference (P>0.05).D total antioxidant capacity was significantly higher than the A,C group(P<0.01),A, B,C group no significant difference(P>0.05).3.4 The Exhaustive time,D rats exhaustive time compared with A,B were significantly increased(P<0.01).B Group A Group to extend the comparison(P<0.05),D group compared to extend the C group(P<0.05).4 Conclusions:4.1 Eight weeks of aerobic endurance training to improve the state of each rat myocardial tissue antioxidant capacity,and enhance enzymatic system to the protective effects of myocardial cells of cardiac tissue cells play a good protection.4.2 Proanthocyanidins reduce free radical scavenging active lipid peroxidation showed a greater advantage,and organizations can improve myocardial antioxidant capacity is conducive to the protection of the myocardium.4.3 Aerobic endurance training in the use of the Joint proanthocyanidins can be an effective way to remove free radicals in myocardial tissue production,lipid peroxidation inhibition, extension of time for rat movement and improve exercise capacity.
