天然胡萝卜素提取及制备相关产品的工艺技术研究

【摘要】 β-胡萝卜素普遍存在于有色蔬菜和水果之中。其在胡萝卜中有较高的含量。β-胡萝卜素是一种抗氧化剂,具有解毒作用,是维护人体健康不可缺少的营养素,在抗癌、预防心血管疾病、白内障及抗氧化上有显著的功能,并进而防止老化和衰老引起的多种退化性疾病。研究立足于我国的资源优势和生产实际情况,针对目前世界市场上对天然β-胡萝卜素不断增大的需求量以及现代人群对营养和保健方面的需要来进行天然β-胡萝卜素提取及相关应用产品的研究开发。进行了如下研究:1.研究建立超声强化样品前处理,RP-HPLC检测β-胡萝卜素的方法体系。色谱条件为色谱柱:(Inertsil ODS-3,C18反向柱,4.6mm×250mm,5μm);流动相:甲醇:氯仿=65:35(v:v);紫外检测器波长:450nm;流速:1.2ml/mim;柱温:30℃;进样量:10μl。并根据均匀试验,得出较优样品检测提取工艺参数为:X1(液固比)=15(v:w),X2(提取时间)=30min,X3(超声功率)=90w,X4(提取温度)=40℃,X5(提取级数)=3次。2研究以胡萝卜干粉为原料,采用超声强化以无毒安全、溶解β-胡萝卜素的能力较好无水乙醇作为溶剂提取醇溶β-胡萝卜素,通过通用旋转正交试验法得出提取最佳工艺条件:X1(提取液固比):30(v:w),X2(提取时间)=45min,X3(超声提取功率)=60w,X4(提取温度)=35℃。开辟了一种简单易行的β-胡萝卜素溶剂提取的新途径。3将天然β-胡萝卜素微胶囊化保持胡萝卜素的固有特性,增强其稳定性克服其极易遭到破坏的缺点,同时弥补了其在水溶液中难于分散的不足。对微胶囊芯壁材制备进行研究,采用喷雾干燥法制备胡萝卜素微胶囊产品。进行均匀试验确定产品工艺和配方为:X1(糊精:麦芽糖浆)=8:2(w:w),X2(芯材:壁材)=4:6(v:w),X3(壁材:水)=1:2.5(w:v),X4(进风口温度)=200℃。并对胡萝卜素微胶囊产品稳定性和品质进行了研究。4研究利用天然β-胡萝卜素微胶囊粉为基质制成适用于儿童、老年人和不能吞服固体制剂的患者食用的口服泡腾片。通过正交感官试验确定泡腾片的工艺配方为:X1(微胶囊粉添加量)=25%,X2[酸源(柠檬酸:苹果酸=1:1)添加量]=35%,X3[碱源(NaHCO3:Na2CO3=1:9)添加量]=20%。同时对产品质量进行了研究。

【Abstract】 β-carotene has widespread in the color vegetable and fruits,especially in carrot. Theβ-carotene are a kind of anti-oxidizing agent,having the function of counteract poison,and a indispensability nourishment that supports the healthy of human body.It also has the function of anti-cancer,preventing cardiovascular disease and anticaducityThe research is having a foothold in actual circumstance of the resources advantage and productions of the country.The research aim at the world to carry on naturalβ-carotene’s extraction and develop the related applied product to demand the modern crowds to nourishment and health care and to demand enlarge continuously market of the naturalβ-carotene currently.The study as follows:1.Study to build up the method:Ultrasonic Extraction to enhance the sample handle,The RP-HPLC examines the method system of theβ-carotene.The HPLC system was consisted of Inertsil ODS-3,C18(4.6 mm×250mm,5μm) and UV detector set at 450nm.The mobile phase:V(CH3OH):V(CHCl3)=65:35,Flow rate:1.2 mL/ min.Temperature of Chromatography:30℃and Inject volume:10μl.The optimum condition has been obtained by Uniform design method as follow:X1(Ratio of solution and solid)=15:1(v:w),X2(Extract time)=30min,X3(The power of ultrasonic)= 90w,X4 (Extract temperature)=40℃,X5(Extract degree)=3 times.2.Study the carrot powder for raw material,Distill theβ-carotene with ethanol which are innocuity and dissolve better.The optimum condition has been obtained by Universal rotary orthogonal experimental design method as follow.X1(Ratio of solution and solid)=30:1(v:w),X2(Extract time)=45min,X3(The power of ultrasonic)= 60w,X4(Extract temperature)=45℃.Develop a new path Extractβ-carotene. 3.The microcapsule of naturalβ-carotene not only keep the carotene but also Strengthen its stability overcome the destructive factor.At the same time to make up for the shortage of difficult dispersion in the aqueous solution.Study on the wall and core materials of microcapsule.Adopt the sprays-drying method to product carotene microcapsule.The optimum condition has been obtained by Uniform design method as follow:X1(The dextrin:The maltose)=8:2(w:w),X2(Corematerials:Wall materials)=4:6(v:w),X3(Wall materials:Water)=1:2.5(w:v),X4(The temperature of entrance)=200℃.Carry on the research to the stability and quality of carotene microcapsule product.4.Study to make use of the microcapsule powder of naturalβ-carotene to make into the Oral effervescent tablets aiming at the child,old people and the person can’t swallow Adopt orthogonal design method to optimum technology condition as follows:X1(The additive of microcapsule powder)=25%,X2[The acid(Citric acid:Malic acid)=1:1]=35%,X3[The alkali(NaHCO3:Na2CO3)=1:9]=20%.Carry on the research to the product quantity at the same time.
