川芎及其主要成分和达乌尔黄鼠冬眠期抗废用性肌萎缩的机制研究

【摘要】 研究目的:1.对川芎及川芎中起活血作用的两种主要药效成分(阿魏酸钠和川芎嗪)对后肢去负荷大鼠比目鱼肌萎缩的影响与作用机制进行初步研究。2.对达乌尔黄鼠冬眠期抗废用性肌萎缩的可能机制进行初步探讨。研究方法:1.尾部悬吊法建立大鼠废用性肌萎缩模型,利用免疫组织化学、血液流变学和梭内外肌纤维超微结构三个方面的研究方法观察药物对比目鱼肌的影响。2.利用血液粘度计和电子显微镜观察并研究达乌尔黄鼠血流变特性及冬眠对梭内外肌纤维超微结构的影响。研究结果:1.与后肢去负荷大鼠相比1)高剂量的阿魏酸钠和川芎嗪使比目鱼肌Ⅰ型肌纤维横截面积分别增加了37.3%和39.4%(P<0.05);2)三种药物均能明显抑制梭外肌纤维MHCⅡ表达水平的升高(P<0.01);3)使肌梭内核袋2纤维MHC的表达由阳性转变为阴性;4)能明显降低低切变率下的全血粘度;5)对梭内外肌纤维超微结构的改变有明显的抑制作用。2.1)冬眠中黄鼠全血粘度较活跃期升高,出眠时下降;2)尾部悬吊对黄鼠的血流变特性无明显改变:3)梭内外肌纤维超微结构在冬眠中无较大改变,出眠时较冬眠中加重。研究结论:1.川芎及其两种主要药效成分阿魏酸钠与川芎嗪均能不同程度地对抗废用性肌萎缩的发生,以高剂量的阿魏酸钠和川芎嗪药效最为明显。2.黄鼠血流变特性和梭内外肌纤维超微结构的改变与其生理特性相关,这可能也是其冬眠过程中抗肌萎缩的原因之一。

【Abstract】 Objective:1. To study the effects and their mechanism of Ligusticum chuanxiong and its two components (Sodium Ferulate and Ligustrazine as main efficacy components in Ligusticum chuanxiong for invigorating blood circulation) on muscle atrophy in a hind limb unloaded rat model.2. To study the ground squirrel’s possible mechanisms of anti-muscle atrophy during hibernation.Methods:1. The tail-suspended rats were subjected to a 14-days disuse, immunohistochemic, hemorheology property and muscle fiber ultrastructure in/out muscle spindle were used to study the effects of medicines on soleus muscle.2. Blood viscometer and Electron microscope were used to investigate the ground squirrel’s hemorheology property and muscle fiber ultrastructure in/out muscle spindle during hibernation.Results:1. Compared with HLU+W, 1) the CSA of soleus type I fibers in HLU+SfH and HLU+TmpH increased by 37.3% and 39.4% respectively (P<0.05); 2) expression level of MHCII were inhibited in all treatment groups (P<0.01); 3) expression of MHC in nuclear bag 2 fiber were altered from positive to negative; 4) the blood viscosity in low shear rates decreased obviously (P<0.001), even near to CON’s; 5) and show the inhibited effect on change of muscle fiber ultrastructure in/out muscle spindle.2. 1) Compare with Aut-A, whole blood viscosity increase during hibernation and decrease after hibernation. 2) Hemorheology property have no significant change after hindlimb-unloading. 3) Muscle fiber ultrastructure in/out muscle spindle have no significant change during hibernation, and after hibernation it worse than H-m. Conclusion:1. Ligusticum chuanxiong and its two main efficacy components (Sodium Ferulate and Ligustrazine) can prevent soleus atrophy induced by disuse, and Sodium Ferulate and Ligustrazine in high dose showed most efficacy.2. Hemorheology property and muscle fiber ultrastructure in/out muscle spindle have relationship with physiological property of ground squirrel, this may be one of the reason that anti-muscle atrophy during hibernation.
