壮瑶五验方抗流感病毒活性研究

【摘要】 目的:对壮瑶五验方的水提物进行抗流感病毒、抗菌、消炎、解热、镇痛方面的研究。方法:(1)用水提醇沉法制备壮瑶五验方的水提物。(2)采用细胞病变效应(CPE)法初步筛选出具有抗流感病毒活性的水提物。(3)采用M-H琼脂平板法检测各验方水提物对几种常见菌及其临床分离菌的最低抑菌浓度(MIC)。(4)采用二甲苯致鼠耳肿胀法对各验方水提物进行抗炎作用的研究。(5)采用干酵母致大鼠发热法对各验方水提物进行解热作用的研究。(6)采用醋酸致小鼠扭体法对各验方水提物进行镇痛作用的研究。结果:(1)制得五验方的水提物(浸膏)。(2)CPE法结果显示,处方1、2、5对流感甲型病毒(粤防72-243)引起的细胞病变有一定的抑制作用,IC50分别为10.69mg生药/ml、3.74mg生药/ml和8.42mg生药/ml,TI分别>1.73、2.32和2.32。(3)体外抗菌试验结果显示,处方1、2、4、5对肺炎链球菌的标准菌ATCC49619及其三个临床分离菌5-18(PRSP)、6-13(PRSP)和9757均有一定的抑菌作用,MIC分别为125mg生药/ml、125~250 mg生药/ml、62.5~250 mg生药/ml和250~500 mg生药/ml。(4)五验方对二甲苯所致小鼠耳肿胀均有一定的抑制作用,其中处方1、2大剂量组的作用尤其显著。(5)处方1、2、3、4、5能抑制于酵母所致的大鼠体温升高。(6)处方1、3、5大剂量组能明显抑制醋酸引起的小鼠扭体反应。结论:壮瑶五验方处方1、2、5有一定的抗流感病毒、抗菌、消炎、解热、镇痛,有深入开发研究价值。

【Abstract】 ObjectiveAnti-viral, anti-bacterial, anti-inflammatory, analgesic and antipyretic effects ofextracts of five formula from Yao and Zhuang minority were investigated.Methods(1)Five formulas from Yao and Zhuang minority were decocted. Ethanol was added tothe decoctions and the dissoluble parts were collected and concentrated.(2)The anti-influenza-viral activities of the extracts were tested by cytopathic effectassay (CPE).(3) The minimal inhibitory concentraction (MIC) of the extracts against some common bacteria and theirs clinical isolates of bacteria were tested by stardard agar dilution method.(4) Mouse ear swelling induced by xylene was used to observe the anti-inflammatory effect of the extracts.(5)Febrile model induced by injecting yeast subcutaneously was used to observeantipyretic effect of the extracts.h(6)Mouse body twist induced by acetic acid was chosen to observe the analgesic effectof the extracts.Results(1)The extracts were obtained from five formulas of Yao and Zhuang Miniotory(aqueous extracts).(2)The result of CPE shows Formula1,2,5 could inhibit the cytopathic effect induced byinfluenza virus, IC50 were 10.69mg /ml, 3.74mg /ml and 8.42mg /ml, TI were morethan1.73,2.32 and 2.32 respectively.(3)The experiment of antibacterial activity in vitro indicates Formula 1,2,4,5 havecertain antibacterial activity against Steptococcus pneumoniae, MIC were 125mg /ml、125~250 mg/ml, 62.5~250mg/ml and 250~500 mg/ml respectively.(4)The five formulas could lessen mouse ear swelling induced by xylene,but the effectof high-dose group of formula 1,2 were more significant than others.(5)Formula 1,2,3,4,5 could inhibit the increases of animal temperature induced by pyrogen.(6)Formula 1,3,5 could inhibit writhing response induced by acetic acid.ConclusionFormula1,2,5 have certain anti-viral, anti-bacterial, anti-inflammatory, analgesic and antipyretic effects. They are of value for further study and development.
