益精方治疗腺嘌呤法不育症大鼠少、弱精症的实验研究

【摘要】 目的:探讨“益精方”治疗腺嘌呤法不育症大鼠少、弱精症的药效学。方法:本实验采用SPF级Wister成年雄性大鼠60只,将大鼠按体重分区再随机分为5组,即正常对照组(12只)、模型组(12只)、低剂量治疗组(12只)、中剂量治疗组(12只),高剂量治疗组(12只)。除对照组外,其余各组均以腺嘌呤按50mg/100g/d剂量灌胃,连续灌胃10天,雄性大鼠不育症模型复制成功。模型制作结束后高、中、低剂量各治疗组分别以“益精方”不同剂量灌胃治疗,灌胃剂量分别为高剂量治疗组3.4g/100g/d、中剂量治疗组1.7g/100g/d、低剂量治疗组0.85g/100g/d。正常对照组和模型对照组每日均以生理盐水灌胃,灌胃容量为1ml/100g(体重),连续灌胃48天。实验过程中观察大鼠一般状态(如毛色、活动、体态等),测日饮水量及进食量。实验结束后将大鼠全部处死,股动脉取血、分离血清、摘取睾丸、性腺及附属性腺称重,提取附睾精液。测量各组大鼠的血清性腺激素(T)及黄体生成素(LH)水平、附睾精子密度和活力,以及睾丸组织学观察。结果:1.与空白对照组比较,模型组精液密度、精子活率和活力及睾酮水平显著下降。2.与模型组相比,各剂量治疗组精液密度、精子活率和活力及睾酮水平显著明显增加,而各治疗组之间无明显性差别。3.腺嘌呤法不育症大鼠经“益精方”治疗后,使受损的睾丸及附属性腺得到一定程度的恢复。结论:1.“益精方”通过调节睾酮(T)分泌,增加睾丸及附属性腺质量,使生精环境得到改善。能够增加精液密度并提高精子的活率及活力,对少弱精症的治疗有很好的疗效。2.“益精方”不同剂量对腺嘌呤法不育症大鼠少弱精症的改善无明显的量效关系,其中低剂量治疗组疗效较为理想。3.“益精方”治疗腺嘌呤法不育症大鼠少弱精症的机理及作用靶点有待进一步研究。

【Abstract】 Objective To study the curative effects of Chinese herb yijingfang on oligospermia and asthenospermia of infertility rats leaded by adenine and analyze corresponding relationship of the dose and the curative effect.Methods Sixty SPF Wistar mature male rats were divided into five groups by their weights: the normal control group(twelve),the model control group(twelve),the treatment group by lower dose(twelve),the treatment group by middle dose(twelve),the treatment group by higher dose (twelve).Except for the normal control group, all the other four groups were perfused with adenine(50mg/100g/d)once a day for ten days, so we established male infertility models of rats. Then the treatment group by higher ,middle and lower dose were perfused with yijingfang once a day for fourty-eight days, the higher dose is 3.4g/100g/d, the middle dose is 1.7g/100g/d,the lower dose is 0.85g/100g/d. The normal control group and the model control group perfused with 0.9% NaCl solution of equivalent volume once a day for fourty-eight days. The Gavage capacity IS 1 ml/100g (weight).Both of the volume is 2ml/100g(weight). In the course of experiment, observe the normal conditions of the rats(such as the colour of the fur ,the activity ,the posture and so on) ,measure the volume of water and food the rats had everday. Then all the rats were killed, serum were separated from blood of femoral artery, testis, gonad, and subsidiary gonad were weighted, semen were imbibed from epididymis, Pathological histological examination was performed on the testis. Then we measure the level of gonadal hormone and luteinizing hormone in plasma and the semen density and active ability of epididymis, observe the histological change of testis before and after treatment.Results①Compared to the normal control group, the model control group had significantly lower spermatozoal density, vitality , active rate and the level of testosterone.②Compared to model control group, the higher ,middle and lower dose groups had significantly higher spermatozoal density, vitality, active rate, showed the obvious differences ,but there are no obvious differences among the three treatment groups.③With the treatment of Yijingfang ,the damaged testis and the subsidiary gonad got some extent recovery. Conclusion"Yijingfang", by regulating testosterone (T) secretion and increase the quality of testicular and subsidiary gonads,improve the spermatogenic environment. It can increase the density of sperm and increase the rate of live sperm and vitality,have a good effect on the treatment of the oligospermia and asthenospermia .2.There is no obvious dose-effect relationship between different doses of”Yijingfang”and the improvement of infertility rats leaded by adenine.The lower dose treatment group has more desirable effect.3. the mechanism and action target of Yijingfang treatment in oligospermia and asthenospermia of infertility rats leaded by adenine is still needed for further research.
