桃核承气汤对蓄血证大鼠血管MMP-2,TIMP-2基因表达的影响

【摘要】 目的观察桃核承气汤(Taohechengqitang,THCQT)对实验性蓄血证大鼠血管基质金属蛋白酶-2(Matrix Metalloproteinase-2,MMP-2)及金属蛋白酶2组织抑制剂(TissueInhibitor of Metalloproteinase-2,TIMP-2)基因表达的影响,探讨桃核承气汤的作用机制。方法采用大肠杆菌内毒素建立蓄血证大鼠模型,大鼠造模后灌服桃核承气汤为药物组,取出空白对照组、模型组、药物组等各组大鼠颈总动脉,应用半定量RT—PCR的方法检测各组MMP-2,TIMP-2基因的表达,分析各组基因表达的变化。结果与空白对照组相比,模型组MMP-2基因表达明显增加,TIMP-2的表达减少(均为P<0.01);与模型组相比,药物组减少了MMP-2基因的表达,增加TIMP-2的表达(均为P<0.01)。结论桃核承气汤能改变实验性蓄血证大鼠动脉壁MMP-2、TIMP-2基因的表达,这可能是桃核承气汤治疗诸多疾病的机制之一。

【Abstract】 Objective To investigate the effects of Taohechengqitang(THCQT) on the gene expression of Matrix Metalloproteinase-2(MMP-2) and Tissue Inhibitor of Metalloproteinase-2(TIMP-2)in artery vessel of experimental Xuxuezheng rats.Methods The model of Xuxuezheng was established by Escherichia Coli endotoxin.Half quantitative reverse transcription polymerase chain reaction(RT-PCR) was used to detect MMP-2 and TIMP-2 gene expression in artery vessel.The changes of gene expression of MMP-2 and TIMP-2 in artery vessels in each group were analyzed.Results MMP-2 gene expression levels in the model group were significantly higher than those of control group(P<0.01),and TIMP-2 gene expression levels were significantly lower (P<0.01).While compared with model group,MMP-2 gene expression levels in the THCQT group were lower(P<0.01),and TIMP-2 gene expression levels were higher(P<0.01).Conclusion THCQT can change the gene expression of MMP-2 and TIMP-2 in artery vessels of experimental Xuxuezheng rats,which may account for the effects of THCQT.
