青紫薯色素毒理学及抗肿瘤活性研究

【摘要】 目的对青紫薯色素的安全性进行评价,并初步探讨其抗肿瘤活性。为青紫薯色素的研发提供实验依掘。方法通过急性毒性实验、Ames试验、小鼠骨髓微核实验、CHL染色体畸变实验和亚慢性毒性实验对青紫薯色素进行安全性评价。复制肉瘤S180小鼠和肝癌H22小鼠模型观察青紫薯色素的抗肿瘤活性。结果1、小鼠灌胃给青紫薯色素一天共21.7g/kg.bw,观察14天未见中毒体征,体重增长正常,根据急性毒性分级确定青紫薯色素为实际无毒物。2、青紫薯色素各剂量组回变菌落数在±S9条件下均未超过阴性对照组回变菌落数的二倍,办无剂量-反应关系。结果说明青紫薯色素0.2~5000μg/皿浓度范围内对四种测试菌株均无致基因突变作用。3、青紫薯色素各剂量组微核率与阴性对照组比较差异无显著性,且P/N>1,实验结果为阴性,表明青紫薯色素在本实验条件下对小鼠骨髓细胞没有抑制作用和毒性。4、在S9活化和非活化两种测试条件下,青紫薯色素在0.25~1.00g/l浓度范围内,诱发CHL细胞染色体畸变率<5%,实验结果为阴性,说明青紫薯色素无致突变作用。5、在亚慢性毒性实验中,青紫薯色素各剂量组大鼠体重、血液生化、主要脏体比等各项指标均在正常范围内。病理组织学观察,肝、脾、肾、胃、十二指肠、睾丸、卵巢等均未见有意义的病理改变。6、大剂量青紫薯色素对S180小鼠的抑瘤率为29.7、45.0和33.3%,中剂量青紫薯色素抑瘤作用不稳定,小剂量青紫薯色素的抑瘤率在10%以下。在同样剂量下,青紫薯色素对H22小鼠抑瘤效果不稳定。结论青紫薯色素在本实验剂量范围内无明显毒副作用,对小鼠肉瘤S180有抑瘤作用。

【Abstract】 AIM:The aim of the present study was to investigate the toxicity of Ipomoea Batatas Poir Cv Anthocyanins,and further to exam its anti-tumor activity.Therefore,to provide the experimental basis for anti-tumor medicine development.METHODS:By acute toxicity testing,Ames test, micronucleus test,CHL chromosomal aberration test and subchronic toxicity testing,the toxicities of Ipomoea Batatas Poir Cv Anthocyanins were detected.The anti-tumor effects of Ipomoea Batatas Poir Cv Anthocyanins were observed on the well-established S180 tumor mice model and hepatoma H22 mice model.RESULTS:1.Mice were treated by Ipomoea Batatas Poir Cv Anthocyanins(intragastric administration,21.7g/kg.bw).After 14 days,no toxic signs were detected.We concluded that Ipomoea Batatas Poir Cv Anthocyanins has no toxicity according to the acute toxicity classification.2. The reverse strain clones induced by different concentrations of Ipomoea Batatas Poir Cv Anthocyanins were less than twice of the control group.No dose-dependent action was observed.This result indicated that Ipomoea Batatas Poir Cv Anthocyanins in 0.2-5000μg has no gene-mutation effect.3.No significance was observed between the different concentrations of Ipomoea Batatas Poir Cv Anthocyanins treatment group and control group in the bone morrow micronucleus test,which indicated that Ipomoea Batatas Poir Cv Anthocyanins has no inhibitory and toxic action on bone morrow cells.4.The Ipomoea Batatas Poir Cv Anthocyanins induced chromosomal aberration was less than 5%in the CHL chromosomal aberration test,no significant deference was detected,which showed that Ipomoea Batatas Poir Cv Anthocyanins could not induce chromosomal aberration.5.In the subchronic toxicity testing,the body weight,food utilization rate, blood property and the size of viscera were also in the normal range in the Ipomoea Batatas Poir Cv Anthocyanins treatment groups.No pathological changes were observed in the morphology of liver, spleen,kidney,stomach,duodenum,testis and ovary.6.The tumor inhibitory rates in sacinoma S180 mice model were 29.7,45.0 and 33.3%separately after high-dose Ipomoea Batatas Poir Cv Anthocyanins treated.While at the middle-dose of Ipomoea Batatas Poir Cv Anthocyanins,the tumor inhibitory rate was instable.The tumor inhibitory rate was less than 10%after low-dose of Ipomoea Batatas Poir Cv Anthocyanins treated.The tumor inhibitory rate in hepatoma H22 mice model was instable compared with that of S180 tumor mice model.CONCLUSION:Ipomoea Batatas Poir Cv Anthocyanins have no adverse effects in the experimental dosage used in this study,and it could have inhibitory effect on sacinoma S180.
