不同留针时间电针对失眠大鼠的催眠作用及其机理研究

The Hypnogenic Effect of Electroacupuncture with Different Needle-retaining Time on Insomnia Rats and Its Mechanism

【摘要】 目的:1.观察不同留针时间电针对戊巴比妥钠致眠大鼠睡眠时间的影响。2.观察不同留针时间电针对对氯苯丙氨酸(PCPA)所致失眠大鼠下丘脑氨基酸类神经递质谷氨酸(Glu)、γ-氨基丁酸(GABA)及一氧化氮(NO)含量、一氧化氮合酶(NOS)活性的影响,探讨电针治疗失眠的机理。3.探讨不同留针时间电针与失眠疗效的相关性。方法:1.采用测定戊巴比妥钠致眠大鼠睡眠时间的方法,观察对照组、电针一组(10min)、电针二组(20 min)、电针三组(30 min)大鼠睡眠时间的变化。2.将Wistar大鼠36只随机分为6组:对照组、模型组、安定组、电针一组、电针二组、电针三组,每组6只。腹腔注射PCPA建立失眠大鼠模型,电针神门和三阴交穴各10 min、20 min、30 min,用离子色谱仪检测大鼠下丘脑Glu、GABA的含量变化,采用化学比色法检测NO含量及NOS活性的变化。结果:1.三组电针组与对照组比较,戊巴比妥钠致眠大鼠的睡眠时间明显延长(P<0.05,P<0.01)。2.大鼠腹腔注射PCPA后,正常的睡眠-觉醒周期消失,三组电针和安定治疗后此周期基本恢复。3.PCPA所致失眠大鼠下丘脑Glu含量、Glu/GABA比值、NO含量及NOS活性显著高于对照组(P<0.01),而GABA含量变化与对照组比较无显著差异(P>0.05)。4.安定治疗后,PCPA所致失眠大鼠下丘脑Glu含量、GABA含量、Glu/GABA比值、NO含量及NOS活性与模型组比较均无明显变化(P>0.05)。5.电针治疗后,PCPA所致失眠大鼠下丘脑Glu含量、Glu/GABA比值显著降低,与模型组、安定组比较均差异显著(P<0.01);GABA含量均升高,但电针一组与模型组、安定组比较均无显著性差异(P>0.05),而电针二组、电针三组与模型组、安定组相比均有显著差异(P<0.05,P<0.01);NO含量及NOS活性显著降低,电针一组与模型组比较差异明显(P<0.05),与安定组比较无明显差异(P>0.05),电针二组、电针三组与模型组、安定组比较均差异明显(P<0.05,P<0.01)。6.三组电针间,电针二组降低Glu含量、Glu/GABA比值、NO含量、NOS活性及升高GABA含量作用均优于电针一组(P<0.05,P<0.01),电针三组与电针一组、电针二组比较均无显著性差异(P>0.05)。结论:1.电针具有镇静催眠作用。2.电针及安定治疗均可使PCPA所致失眠大鼠的睡眠-觉醒周期恢复,电针调节Glu、GABA、NO、NOS作用优于安定,电针可能是通过降低失眠大鼠下丘脑Glu含量、Glu/GABA比值、NO含量、NOS活性,升高GABA含量达到治疗失眠的目的。3.电针20 min治疗失眠作用优于10 min、30 min。

【Abstract】 Objective1.To observe the effects of different needle retaining time of electroacupuncture on the sleep time of Pentobarbital sodium rats.2.To observe the effects of electroacupuncture in different needle retaining time on the contents of Amino Acid Neurotransmitter such as Glutamic acid(Glu),γ-Aminobutyric acid(GABA), and Nitrogen monoxidum(NO) content,Ntric oxide synthase(NOS) activity in the hypothalamus of insomnia rats caused by DL-P-Chlorophenylalanine(PCPA) to investigate the mechanism of electroacupuncture treatment of insomnia.3.To explore the relation between time of needle retaining in electroacupuncture treatment and the effect of insomnia rats.Methods1.The sleep time of Pentobarbital sodium rats was recorded to observe the changes of sleep time of rats in control group,electroacupuncture groupⅠ(10 min),electroacupuncture groupⅡ(20 min) and electroacupuncture groupⅢ(30 min).2.Totally 36 female Wistar rats were divided randomly into 6 groups with 6 rats in each group: the control group,the model group,the diazepam group,electroacupuncture groupⅠ, electroacupuncture groupⅡand electroacupuncture groupⅢ.The model of insomnia rats were established by PCPA treatment i.p and the acupoints of Shenmen(HT7) and Sanyinjiao(SP6) were chosed to be acupunctured by needles.The change of Glu,GABA,NO and NOS in the hypothalamus of rats were observed by Ion Chromatograph and chemical chromatometry.Results1.Compared with the control group,the sleep time of Pentobarbital sodium rats was obviously prolonged in three electroacupuncture treatment groups(P<0.05,P<0.01).2.After PCPA treatment,the normal sleep-wakefulness cycle of rats disappeared;but both of electroacupuncture and Diazepam could recover the cycle.3.After PCPA treatment,the contents of Glu,Glu/GABA,NO and NOS activity in the hypothalamus of rats obviously increased with comparison to control group(P<0.01),but the content of GABA did not change much compared with control group(P>0.05).4.After Diazepam administration,the contents of Glu,GABA,Glu/GABA,NO and the activity of NOS in the hypothalamus of insomnia rats caused by PCPA had no obvious change with comparison to the model group(P>0.05).5.After electroacupuncture treatment,the contents of Glu and GIu/GABA decreased obviously in contrast with the model group and the diazepam group(P<0.01);while that of GABA increased,but electroacupuncture groupⅠhad no obvious change with comparison to the model group and the diazepam group(P>0.05),electroacupuncture groupⅡand electroacupuncture groupⅢincreased obviously in contrast with the model group and the diazepam group(P<0.05, P<0.01).At the same time,the content of NO and the activity of NOS decreased greatly, electroacupuncture groupⅠchanged obviously in contrast with the model group(P<0.05),but had no obvious change with comparison to the diazepam group(P>0.05),electroacupuncture groupⅡand electroacupuncture groupⅢchanged obviously in contrast with the model group and the diazepam group(P<0.05,P<0.01).6.Among the three electroacupuncture groups,groupⅡhad a better effect on decreasing the contents of Glu,Glu/GABA,NO and the activity of NOS,and increasing the content of GABA than that of groupⅠ(P<0.05,P<0.01).There was no significant difference of the effect between the groupⅢand the other group(P>0.05).Conclusion1.Electroacupuncture has mitigated and hypnotic effect.2.Both of electroacupuncture and Diazepam can recover the sleep-wakefulness cycle of insomnia rats caused by PCPA,but electroacupuncture is superior to Diazepam to regulate on Amino Acid Neurotransmitter such as Glu,GABA and NO,NOS.The mechanism of electroacupuncture in treating insomnia is decreasing Glu,Glu/GABA,NO,NOS and increasing GABA in the hypothalamus of insomnia rats.3.Electroacupuncture groupⅡ(20 min) has a better effect than groupⅠ(10 min) and groupⅢ(30 min) on treating insomnia.
