环糊精及其衍生物对药物等有机分子的识别与选择性提取

【摘要】 β-环糊精能选择性地与各种有机、无机、生物分子形成主-客体或超分子配合物,常被用作分子主体来研究其对各种有机、无机、生物分子的分子识别作用。近年来,具有良好水溶性的2-O-(2-羟基乙基)-β-环糊精、甲基-β-环糊精和2-O-(2-羟基丙基)-β-环糊精等环糊精衍生物,已被广泛地研究对于药物的包结复合,以提高药物的溶解度和生物利用度。本论文主要研究环糊精及其新型衍生物6-O-聚乳酸-β-环糊精对于药物等有机分子的识别和选择性提取。本论文共分为两大部分:第一部分通过紫外法和相溶解度法研究了β-环糊精及其衍生物(2-羟丙基-β-环糊精、6-羟丙基-β-环糊精和6-聚乳酸-β-环糊精)对客体分子的包结复合,测定了包结常数,从理论上解释了不同β-环糊精衍生物对客体分子的识别与增溶;第二部分研究了2-羟丙基-β-环糊精对于复杂体系(如木瓜叶黄酮和有机小分子)的选择性提取。本论文分为四章:一、通过紫外法研究了新型环糊精衍生物对于难溶药物(芦丁、阿莫西林)的包合性能,求出β-环糊精衍生物对客体的包结常数,对比不同β-环糊精衍生物对同种客体的分子识别能力,为解释难溶药物的增溶现象提供了理论依据,具有一定的理论和应用意义;这也充分验证了β-环糊精衍生物有很好的分子识别能力。人工合成的植物生长调节剂萘乙酸(NAA)和6-苄氨基嘌呤(6-BA)具有广泛的用途,但储存时间短,若选择一种便于保护的稳定剂,以增加药剂存放时间,则会保证其应用价值的最大实现。本论文研究水溶液中环糊精及其衍生物与萘乙酸、6-苄氨基嘌呤的包合反应,这些结果对研究环糊精溶液中的包络平衡及萘乙酸、6-苄氨基嘌呤的控释有重要的参考价值。二、通过相溶解度法研究不同类型的环糊精衍生物对难溶药物(芦丁、阿莫西林)和植物生长调节剂(萘乙酸和6-苄氨基嘌呤)的包合过程,求出β-环糊精衍生物对客体的包结常数,为难溶药物的增溶和控释提供理论依据。三、结合一和二结果可知,2-羟丙基-β-环糊精对芦丁有较强的识别能力,尝试提取木瓜叶中黄酮成分的新方法,创新性地利用2-羟丙基-β-环糊精溶液作为木瓜叶中黄酮提取剂,分别了讨论不同溶剂、不同温度、不同2-HP-β-CD溶液浓度和不同料液比条件对提取效果的影响,得到最佳提取条件为:1mmol/L2-HP-β-CD水溶液,料液比1:20,60℃水浴加热3h,为木瓜叶中黄酮成分的提取工艺提供理论依据。四、利用2-羟丙基-β-环糊精溶液作为萃取剂,对苯-甲苯体系、苯-苯酚-苯甲醛体系、苯-苯胺-苯酚-苄胺体系和苯-苯酚-苯甲酸体系进行萃取分离,对比研究了2-羟丙基-β-环糊精对结构相似的有机小分子物质的识别能力。

【Abstract】 Cyclodextrins are widely used in the field of molecular recognition.It could form inclusion compounds with organic compounds,inorganic compounds and biology compounds because of supermolecular interaction.Recently,β-cyclodextrin derivatives such as 2-O-(2-hydroxyethyl-β-cyclodextrin)and 2-O-(2-hydroxylpropyl -β-cyclodextrin)and methyl-β-cyclodextrin which have good solubility are widely used in drug complex for drug carrier and recognition,increasing drug solubility and bioavibility.My work focus on the recognition and selective extraction of cyclodextrin and its derivatives 2-O-(2-hydroxypropyl)-β-cyclodextrin(2-HP-β-CD), 6-O-(2-hydroxypropyl)-β-cyclodextrin(6-HP-β-CD),6-polylacticacid-β-cyclodextrin (6-PL-β-CD).This work has two parts.One is the recognition process and enhanced solubility to guest detected by UV-Vis spectroscopy and the other is the selective separation of 2-HP-β-CD to the flavones of papaya leaves and small molecules.The content of the paper was showed as follows:1.The inclusion constant ofβ-CD derivates to pharmacy with low solubility (rutin,amoxicillin)for molecule recognition have been calculated.Comparing the different molecules recognition ability,it was found that theβ-CD derivates showed strong recognition ability which could be used as good efficient enhanced regent for rutin and amoxicillin.Naphthylacetic acid(NAA)and 6-benzyl aminopurine(6-BA) have been used in many areas as good artificial plant growth regulators.But the time for storing is short.The vitality could be found if we can find a good storage agent. This paper focuses on the inclusion behaviors ofβ-CD derivates to NAA and 6-BA.It would provide some useful information on controlling and releasing NAA and 6-BA.2.The inclusion constant ofβ-CD to low solubility pharmacy(such as rutin, amoxicillin,NAA,6-BA)has been studied by phase-solubility method,which has provided a valuable reference for enhanced solubility and controlled drug release.3.The trials for extracting the flavone from the papaya leaves have been directed. The influences of solvent,temperature,and concentration of the host-guest molar ratio were discussed.The best condition was as follows:2-HP-β-CD(lmmol/l),raw materials 1:20,60℃for 3h.4.The 2-HP-β-CD was used as extract agent,the separation of benzene-toluene system,benzene-phenol-benzaldehyde system,benzene-aniline-phenol-benzylamine system and benzene-phenol-benzoic acid system have been directed.Different recognition ability of 2-HP-β-CD to small organic molecule was discussed.
