丹川芪颗粒对急性心肌缺血大鼠血清降钙素基因相关肽水平的影响

【摘要】 目的:通过观察丹川芪颗粒对急性心肌缺血大鼠血清降钙素基因相关肽水平的影响,探讨丹川芪颗粒治疗急性心肌缺血的作用机制,从而为丹川芪颗粒在临床上的应用提供科学的理论依据。方法:将Wistar大鼠50只,雌雄各半,随机分为五组,分别为丹川芪颗粒高、低剂量组,通心络对照组、模型对照组和正常对照组,每组各10只。各组分别于术前24h,12h及1/2h各灌胃1次,末次灌胃30min后进行实验。除正常对照组以外,其余各组大鼠分别采用结扎大鼠冠状动脉的方法建立急性心肌缺血的模型,造模成功30min后于门静脉取血,测定血清降钙素基因相关肽含量。结果:1.丹川芪颗粒高、低剂量组、通心络对照组和模型对照组与正常对照组相比血清降钙素基因相关肽的水平均降低,有显著性差异(P<0.05);2.丹川芪颗粒高、低剂量组、通心络对照组与模型对照组相比血清降钙素基因相关肽的水平均升高,有显著性差异(P<0.05);3.丹川芪高、低剂量组与通心络对照组相比较,血清CGRP水平无显著性差异(P>0.05)。4.丹川芪高剂量组与丹川芪低剂量组相比较,血清CGRP水平无显著性差异(P>0.05)。结论:丹川芪颗粒对急性心肌缺血的治疗机制可能与升高血清降钙素基因相关肽的水平有关,并由此推测该药治疗急性心肌缺血的疗效和机制与通心络胶囊相似。

【Abstract】 Objective:To observe the effect of Danchuanqi Granulas on calcitonin gene-related peptide in rats with acute myocardial ischemia and discuss the mechamism of Danchuanqi Granulas on treating the acute myocardial ischemia,providing theory for clinical application of Danchuanqi Granulas.Method:We divided the fifty wistar rats(male:female 1:1)into five groups.The high dose group and low dose group of Danchuanqi Granulas, comparison group of Tongxinluo,model comparison group and normal comparison group,ten rats a group.Every group had oral administration before the operated time of twenty four hours,twelve hours and half hour respectively,and gave operation on thirty minutes after the last oral administration.Excluding the normal comparison group we set up the acute myocardial ischemia model by the method of ligating the coronary artery of the rats on other groups,took blood from portal vein after thirty minutes on the base of the successful making model,determined the content of calciatonin gene-related peptide.Result:1.Compared with the normal comparison group,the level of calcitonin gene-related peptide in the high dose group and low dose group of Danchuanqi Granulas,comparison group of Tongxinluo and model comparison group is averagely degraded,the difference is significant(P<0.05);2.Compared with the model comparison group,the level of Danchuanqi Granulas, comparison group of Tongxinluo is averagely heightened,the difference is significant(P<0.05);3.The calcitonin gene-related peptide level in serum in the high dose group and low dose group of Danchuanqi Granulas has no significant difference with Tongxinluo comparison group(P>0.05); 4.Comparing with the low dose group of Danchuanqi Granulas,the level of calcitonin gene-related peptide in serum in the high dose group of Danchuanqi Granulas has no significant difference.(P>0.05)Conclusion:The mechanism of Danchuanqi Granulas on treating the acute myocardial ischemia is related to the level of calcitonin gene-related peptide.According to the experimental result,we can infer that Danchuanqi Granulas have the similar effect and mechanism with o Tongxinluo capsule on treating acute myocardial ischemia.
