新生大鼠海马脑片OGD模型中HO-1对NGB的影响

【摘要】 目的:通过观察血红素加氧酶(heme oxygenase, HO)对脑红蛋白(Neuroglobin,NGB)的影响,探索NGB的调控机制。方法:体外培养新生8-10天SD大鼠海马脑片,两周后应用HO-1促进剂Hemin和抑制剂Znpp干预后建立OGD模型。采用HE、尼氏染色和免疫组化方法观察海马脑片OGD模型中神经元和星形胶质细胞的状态及HO-1和NGB表达的变化。结果:①在加入Hemin后建立的海马脑片OGD模型中,脑片HE及尼氏染色显示神经元和星形胶质细胞与正常培养的海马脑片比较变化不大,仅有轻微的水肿。脑片免疫组化结果显示HO-1和NGB的表达均较正常培养建立的OGD模型中增加。②在加入Znpp后建立的海马脑片OGD模型中,脑片HE及尼氏染色显示神经元和星形胶质细胞结构模糊,存在广泛水肿、变性、坏死。脑片免疫组化结果显示HO-1和NGB的表达均较正常培养建立OGD模型中下降。结论: HO-1和NGB呈同步变化,提示两者之内可能存在相关关系。NGB对神经元和星形胶质细胞具有保护作用。

【Abstract】 Objective: By observing the effect of heme oxygenase on neuroglobin, explore the regulation mechanism of NGB .Methods: Firstly, cultivate hippocampal slices from 8~10-day-old rats in vitro. After two weeks, add accelerant Hemin and inhibitor Znpp of HO-1into hippocampal slices respectively. Then, construct OGD model. By HE, Nissl’s staining and immuno histochemistry, observe the state of neuron and astrocyte and the change of HO-1 expression and NGB expression.Result:①In the model of OGD made by adding accelerant Hemin,it is observed by HE and Nissl’s staining that neuron and astrocyte are not different from the cells cultivated normally, except for slight edema. However, the results of immuno histochemistry of hippocampal brain slice show that there are more HO-1 expression and NGB expression than those of hippocampal brain slice cultivated normally.②In the model of OGD made by adding inhibitor Znpp, it is observed by HE and Nissl’s staining that some neuron and astrocyte have vague structure , some are general edema, some are apomorphosis or necrosis. However, the results of immuno histochemistry of hippocampal brain slice show that there are less HO-1 expression and NGB expression than those of hippocampal brain slice cultivated normally.Conclusion: The isochronous changes of HO-1 and NGB show that there is probably relative relation between HO-1 and NGB. NGB can protect neuron and astrocyte.
