PECAM-1在口腔疣状癌表达研究

【摘要】 目的:研究血小板-内皮细胞粘附分子(platelet-endothelial celladhesion molecule-1,PECAM-1)在口腔疣状癌(Oral verrucouscarcinoma,OVC)和口腔鳞癌(oral squamous cell carcinoma,OSCC)中的表达,探讨其在OVC和OSCC发生发展中的作用,对基因芯片结果进行验证。方法:取9例外生型口腔疣状癌、6例囊肿/浸润型口腔疣状癌、15例高中分化鳞癌、15例低分化鳞癌,应用半定量逆转录多聚酶链式反应(RT-PCR)方法检测OVC和OSCC中PECAM-1 mRNA的表达水平,应用免疫组织化学法检测上述标本中PECAM-1的表达及分布,判断差异。结果:1、PECAM-1的IHC实验a、四种癌组织中PECAM-1的微血管密度(MVD)均高于正常粘膜组织(P<0.05)。b、口腔鳞癌中,高/中分化鳞癌与低分化鳞癌的MVD有显著性差异(P<0.05)。c、外生型疣状癌、高/中分化鳞癌、囊肿/浸润型疣状癌、低分化鳞癌,四组的PECAM-1的MVD依次升高(P<0.05)。2、PECAM-1的RT-PCR实验a、外生型疣状癌、高/中分化鳞癌、囊肿/浸润型疣状癌、低分化鳞癌四组中的PECAM-1 mRNA表达明显高于相应正常组(P<0.05)。b、高/中分化鳞癌与低分化鳞癌比较:前者PECAM-1 mRNA的表达水平低于后者,二者差异显著(P<0.05)。c、外生型疣状癌、高/中分化鳞癌、囊肿/浸润型疣状癌、低分化鳞癌,四组的PECAM-1 mRNA的表达依次升高(P<0.05)。结论1.PECAM-1可能在口腔疣状癌和口腔鳞癌发生发展过程中起了重要作用,其中促进肿瘤血管的生成是其主要的发病机制。2.PECAM-1在口腔疣状癌和口腔鳞癌的表达差异,为进一步说明二者不同的生物学行为提供了可靠的依据。3.PECAM-1分别从RT-PCR和免疫组化学层面,说明了囊肿/浸润型口腔疣状癌可能有高于外生型疣状癌以及部分口腔鳞癌的浸润潜能。

【Abstract】 Objective To study the expression the platelets - endothelial cell adhesion molecule(PECAM-1)in oral verrucous carcinoma(OVC)and oral squamous cell carcinoma(OSCC),so as to investigate the role of this gene in the development of OVC and OSCC,and to validate the results of gene chip.Methods 9 cases of exogenic OVC、6 cases of cystoid and infiltrative OVC、15 cases of well/moderately- differentiation OSCC (w/md-OSCC)and 15 cases of poor-differentiation OSCC(pd-OSCC) were selected;two specimens,which were carcinoma tissues and the surround normal tissues,were taken from the same patients.Test the mRNA expression of PECAM-1 gene by reverse transcription polymerase chain reaction(RT-PCR)and check the PECAM-1 protein by immunohistochemistry(IHC)in the above specimens.Results1.PECAM-1 of the IHC testa.The microvessel density(MVD)of PECAM-1 in 2 types OVC and 2 types OSCC was higher than normal tissues(P<0.05).b.There was difference of PECAM-1’s MVD between w/md-OSCC and pd-OSCC(P<0.05). c.The MVD of PECAM-1 was significantly higher in this order:Exogenic verrucous carcinoma,Well differentiated squamous cell carcinoma,Cystoid/infiltrative verrucous carcinoma,Poorly differentiated squamous cell carcinoma.(P<0.05).2.PECAM-1 of the RT-PCR testa.The mRNA expression of PECAM-1 gene was significantly higher in both exogenic OVC and cystoid and infiltrative OVC than in their matched normal tissues(P<0.05);The mRNA expression of PECAM-1 gene was also significantly higher in both w/md-OSCC and pd-OSCC than in their matched normal tissues(P<0.05).b.There was difference in the mRNA expression of PECAM-1 gene between w/md-OSCC and pd-OSCC(P<0.05).c.The mRNA of PECAM-1 was significantly higher in this order:Exogenic verrucous carcinoma,Well differentiated squamous cell carcinoma,Cystoid/infiltrative verrucous carcinoma,Poorly differentiated squamous cell carcinoma.(P<0.05).Conclusion1.PECAM-1 may play an key role in the invade and metastasis of OVC and OSCC,Of which promote the formation of tumor blood vessels is the main mechanism for the pathogenesis. 2.The difference of PECAM-1 in OVC and OSCC may be the gene foundation of invade and metastasis that the potential of OVC is lower than OSCC.3.And it also showed part of cystoid/infiltrative OVC can have a higher potential of invade and metastasis than OSCC.
