组织工程化生物活性骨膜应用于兔腰椎横突间融合的实验研究

【摘要】 组织工程化生物活性骨膜应用于兔腰椎横突间融合的实验研究【目的】评价体外构建组织工程化生物活性骨膜为骨移植替代物应用与兔腰椎横突间融合的效果。【方法】体外构建组织工程化生物活性骨膜植入24只健康成年新西兰大白兔腰椎横突间,每只兔子取三个横突间隙(LeftL4,5,6、RightL4,5,6)植入3种材料(RightL4,5植入诱导后骨髓间充质干细胞复合猪小肠黏膜下层SIS(A组)、RightL5,6(B组)植入无细胞支架即单纯SIS、LeftL5,6植入自体髂骨(C组))。术后12W处死动物进行大体标本、影像学、组织学观察。【结果】大体标本比较A、C两组无统计学意义,均与B组有统计学意义,影像学观察A、C两组上下横突间可见明显有骨小梁通过,B组未见骨密度影。A组Ⅰ-型胶原、骨钙素免疫组织化学染色强阳性,与C组之间有统计学意义,B组未见阳性表达。【结论】利用猪小肠黏膜下层(SIS)复合经诱导后向成骨细胞转化的骨髓间充质干细胞(BMSc)体外构建组织工程化生物活性骨膜是横突间融合骨移植的良好替代物。

【Abstract】 The Empirical Study for Tissue Engineered Biological Activity Periosteum Used in Spinal Fusion of Lumbar Intertransverse Process in Rabbits[Objective]to appraise the effect of out-of-body construction tissue engineered biological for bone transplantation surrogate using and spinal fusion of lumbar intertransverse process in rabbits.[Method]out-of-body construction tissue engineered biological implant in 24 healthy adult New Zealand rabbits of lumbar intertransverse process.Each rabbit is taken three transverse process gaps LeftL4,5,6、RightL4,5,6.implant Three kinds of materials: imbedding RightL4,5the induced bone mesenchymal tissue stem cells combination with the pig’s small intestine submucosa SIS(A);imbedding RightL5,6the cytoskeleton(B);imbedding the LeftL5,6autoallergic flank bone.,Operation the rabbits after 12W,observe the manual palpation,radiography,histology test.[results]manual palpation compared,A,C hasn’t statistically significant,but each other has statistically significant with B.By radiography observation,A,C groups’ lumbar intertransverse has bone trabecula significantly,but B hasn’t bone density.A group I-ollagen,osteocalcin immunohistochemistry dyeing is hadro-masc,which has statistically significant with C,but B group hasn’t masc.[Conclusion]Using SIS to compound induced bone mesenchymal tissue stem cells out-of-body construction tissue engineered biological is good surrogate.
