土壤干旱胁迫对金线兰三种组培苗抗旱性的影响

【摘要】 金线兰是一种珍贵的药用植物,且观赏价值极高,一般生长于高海拔之阴凉地区,性喜冷凉气候、湿润环境,对环境要求很高,加上野生资源稀少,因此发展金线兰人工栽培势在必行。干旱环境很容易造成金线兰不正常生长甚至死亡。因此,本论文以三种来源不同的金线兰组培苗为材料,设置五个土壤干旱梯度(9、13、17、21、25天),以0天为对照,研究三种金线兰组培苗在土壤胁迫下生理响应机制,并分析生理指标变化,采用因子分析法和隶属函数法对金线兰组培苗抗旱性作出评价,为其大量人工栽培提供理论依据和科学指导。试验结果表明:(1)金线兰在土壤干旱逆境胁迫下体内发生了一系列的生理响应机制,12项指标的变化趋势是:RWC、Chl含量、Car含量一直下降;PMP、MDA含量、可溶性糖一直升高;POD、SOD、CAT酶活性、可溶性蛋白含量、脯氨酸都是先升高后下降;APX酶活性则是先下降后升高。指标系数的相关距阵表明各个指标之间都有或大或小的相关性,反映的信息在一定程度上有重叠,因此本论文采用因子分析将12个指标缩减为两个公因子,确定公因子权重,最后进行综合分析。(2)通过因子分析和隶属函数法对金线兰抗旱性评价结果表明,金线兰三种组培苗抗旱性排列顺序为:种子苗>根状茎苗>地上茎苗。这结果可为金线兰的人工栽培起指导作用。

【Abstract】 Anoectochilus roxburghii(Wall.)lind is a rare medicinal herbs and have high enjoy value, usually it existences in place of high altitude ,loving shady and cool climate, and have a strict demand on environment, besides wilding are very exiguous,so it is very important to development cultivation.Drought environment may easily lead to unusual growth even death of A.roxburghii.So this paper taked three different sources of A.roxburghii. tissue cultured seedlings as experimental materials,setting up soil drought stress of five gradients (9、13、17、21、25 days) and 0 day for comparison, to study three kinds of A. roxburghii. tissue cultured seedlings physiological response of drought stress, then measure physiological and biochemical indexes and analyze them,using principal component analysis and subordinate function values analysis to estimate drought resistance of A.roxburghii. tissue cultured seedlings,and provide theory basis and scientific direction for numerous cultivation of A.roxburghii.Research results showd that:( 1 ) Anoectochilus roxburghii(Wall.)lind occurd an array of physiological response under drought stress, change tendency of 12 indexes is: RWC、content of Chl and Car always reduced; PMP、content of MDA and soluble sugar always increased;activity of POD、SOD、CAT、APX and content of proline and soluble protein exhibited up-down trends;APX activity exhibited down-up trends.correlation matrix of every indexes showd that every indexe had more or less correlation,and reflectd repeatd information.So this paper used principal component analysis to reduced 12 indexes for 2 components,and decide component weight,and finally proceed comprehensive analysis.(2)Through the principal component analysis and subordinate function values analysis,the results of evaluation on drought resistance of A.roxburghii shown that the ability of drought resistance of three tissue cultured seedlings of A.roxburghii. on different drought stress: S > R > OS,The result can provide direction for cultivating of A.roxburghii.
