长俊木瓜试管苗前期培养时间、株高和芽数对生根培养的影响及DG增殖方法的研究

【摘要】 木瓜为蔷薇科(Rosaceae),木瓜属(Chaenomeles lindi)植物,全属5种,皱皮木瓜(Chaenomeles speasose)是其中之一。长俊木瓜(Chaenomeles speciosa(sweet) Nakai C.Lagenaria)(chang jun)为皱皮木瓜中的一个优良品种。长俊木瓜的医用、食用及观赏价值已经被广大群众所接受,因此研究其组培快繁不仅有很高的科研价值,还会带来可观的经济价值。本文在前人对长俊木瓜组培快繁研究的基础上,应用适合的生根培养基和培养条件,分析得出长俊木瓜无菌苗生根情况个体差异大的原因为前期培养时间、株高及芽数的不同。因此本文就这3个方面对其对长俊木瓜生根的影响做了具体研究。在长期的组织培养过程中,研究发现了一种新的无菌苗增殖方法——DG增殖方法,本文也对此做了深入研究。具体结论如下:1.前期培养时间、株高及芽数对长俊木瓜无菌苗生根的影响1.1长俊木瓜无菌苗最适合的前期培养时间为6个月。1.2 2cm以下的长俊木瓜无菌苗一个月内不能生根。1.3无菌苗株高越高,芽数越多,长俊木瓜无菌苗生根越有利。2.DG增殖方法的研究2.1 DG增殖方法进行芽诱导,其增殖系数和无菌苗株高变化都大于常规增殖方法,DG增殖方法有利于长俊木瓜无菌苗的增殖。2.2来自DG增殖方法的无菌苗,其生根率和根长都明显好于来自常规增殖方法的无菌苗,来自DG增殖方法的无菌苗生根效果好,且株高越高,芽数越多的无菌苗越有利于生根。2.3 DG增殖方法的无菌苗经切割后,剩余的各种苗态的无菌苗在增殖培养基中,生长状况良好,增殖及生长能力较强。

【Abstract】 Chaenomeles is plant in Rosaceae, Chaenomeles lindi, altogether is five, Chaenomeles speasose is one of them. Chaenomeles speciosa (sweet) Nakai C.Lagenaria (chang jun) is a new good species in Chaenomeles speasose.The medicinal、edible and ornamental value of Chaenomeles speciosa(sweet) Nakai C.Lagenaria (chang jun) is already accepted by most people, so the studying on Chaenomeles speciosa (sweet) Nakai C.Lagenaria (chang jun)’s tissue culture and rapid propagation not only has the researching value but also has the economic value.On the basis of the previous study, I cultured the Chaenomeles speciosa (sweet) Nakai C.Lagenaria (chang jun) on the rooting medium under the feasible culture conditions.I analyzed the reason why the rooting-rates were so different among all the aseptic seedlings under the same conditions.The results of the analysis showed that the difference of proliferation duration, the height and the bud number of aseptic seedlings were the influencing factors.So I studied the concrete effects on rooting with these three aspects.During the long time studying, a new method of proliferation—DG method of proliferation was found.I had done the further study on this method.The main results were as follows:1 The effects of proliferation duration, height and bud number of Chaenomeles speciosa (sweet) Nakai C.Lagenaria (chang Jun)’s aseptic seedling on rooting.1.1 Sixth month was the most suitable time of proliferation duration.1.2 The aseptic seedlings whose heights were lower than 2cm could not be rooting within one month.1.3 The higher of aseptic seedling was and the more bud number of aseptic seedling had the more suitable for rooting.2 The study on DG method of proliferation2.1 The changes of proliferation index and the height of aseptic seedling were more obvious when using DG method of proliferation than traditional method of proliferation.DG method of proliferation was more suitable for proliferation. 2.2 The effects of rooting rate and root-length were better when using DG method of proliferation than traditional method of proliferation.DG method of proliferation was more suitable for rooting, and the higher of aseptic seedling was and the more bud number of aseptic seedling had, the more suitable of rooting.2.3 The aseptic seedlings with different conditions were the cut aseptic seedlings which were cultured on DG method of proliferation were growing well, and had the higher ability of rooting and proliferation.
