维药芹菜根醇提物护肝降脂作用的初步探索

【摘要】 目的:初步探索维药芹菜根醇提物对四氯化碳(CCl4)、D-氨基半乳糖(D-GaIN),扑热息痛(PAR)所致的急性肝损伤的保护作用及对高脂血症大鼠血脂的调节作用。方法:(1)选用Wistar大鼠,分别用CCl4(1ml/kg)、D-GaIN(500mg/kg)进行攻击;选用昆明小鼠,用大剂量PAR(300mg/kg)进行攻击。分别建立以上三种肝损伤模型,观察维药芹菜根醇提物对实验动物血清谷丙转氨酶(ALT)、谷草转氨酶(AST)和肝组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量及病理变化的影响。(2)选用Wistar大鼠,采用灌胃脂肪乳剂的方法建立大鼠高脂血症模型,分别用3g/kg、1.5g/kg剂量的维药芹菜根醇提物连续喂饲大鼠30天,取血清测定大鼠血清总胆固醇(CHO)、甘油三脂(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量。结果:(1)芹菜根醇提物能明显降低CCl4、D-GaIN引起的急性肝损伤动物血清ALT、AST水平,其高剂量能明显提高肝组织SOD活性,降低MDA含量。芹菜根醇提物能明显升高PAR引起的急性肝损伤动物肝组织SOD活性,降低MDA含量,其高剂量能明显降低动物血清ALT、AST水平。肝组织病理变化观察发现芹菜根醇提物能改善CCl4、D-GaIN、PAR所致肝组织的损伤程度。(2)芹菜根醇提物能明显降低高脂血症大鼠血清CHO、TG、LDL-C和MDA含量,增高HDL-C及SOD活性。结论:维药芹菜根醇提物对CCl4、D-GaIN、PAR引起的急性肝损伤有一定的保护作用,并可降低高脂血症大鼠的血脂水平。

【Abstract】 Objective: To observe the effect of aqueous extract of alcohol of the roots of Apium graveolens L on the acute liver injury that induced by carbon tetrachloride (CCl4)、D-Galactasamine (D-GaIN) and paracetamol(PAR). And to observe the influence of aqueous extract of alcohol of the roots of Apium graveolens L on experimental hyperlipidemia of rats. Methods: (1)Experimental hepatitis animal models were established in wistar rats by an single intraperitoneal injection of CCl(41ml/kg)、D-GaIN(500mg/kg),and an single intraperitoneal injection of PAR(300mg/kg )in mice,respectively. Serum Alanine transferase(ALT), Aspartate tranfe- rase(AST), liver Super oxide dismutase (SOD), Malondialdehyde(MDA) were examined. Also histopathological changes in the liver tissue were observed. (2)Set up the rats hyperlipidemia model by ig lipid emulsion, rats that in treatment groups were continually given aqueous extract of alcohol of the roots of Apium graveolens L at dosages of 3g/kg、1.5g/kg separately for 30 days. The serum levels of CHO, TG ,HDL-C, LDL-C, SOD, and MDA of rat were examined after 30 days. Results: (1)Both dosages of aqueous extract of alcohol of the roots of Apium graveolens L significantly decreased serum ALT and AST elevation and the high-dosage increased the SOD activities while lowered MDA content significantly in the CCl4 and D-GaIN induced hepatitis model. Both dosages of aqueous extract of alcohol of the roots of Apium graveolens L increased the SOD activities while lowered MDA content significantly in the PAR induced hepatitis model. And the high-dosage significantly decreased serum ALT and AST elevation. It was concl- uded that the aqueous extract of alcohol of roots of Apium graveolens L possesses a significant hepatoprotection against the three kinds of chemical induced hepatitis. (2)The aqueous extract of alcohol of the roots of Apium graveolens L significantly decreased serum CHO,TG,LDL-C and MDA, increased HDL-C and SOD on experimental hyperlipidemia of rats. Conclusion: The aqueous extract of alcohol of the roots of Apium graveolens L has protective effects for acute liver injury in rats induced by CCl4、D-GalNand PAR. It could reduce the level of blood lipids on experimental hyperlipidemia of rats.
