芪茵颗粒对非酒精性脂肪肝大鼠瘦素及胰岛素抵抗的影响

Experimental Study on Curative Effect and Mechanism of QiYin Granular Formulation on Nonalcoholic Fatty Liver Disease in Rat

【摘要】 目的:观察芪茵颗粒对非酒精性脂肪性肝病(NAFLD)大鼠血清瘦素及胰岛素抵抗指数的影响及对肝脏组织病理学的影响,为进一步探讨脂肪肝的形成机制及芪茵颗粒治疗作用途径,及为临床应用提供较好的理论及实验依据。方法:60只Wistar大鼠雌雄各半,随机分为六组:空白对照组、模型对照组、易善复对照组、芪茵颗粒低剂量、芪茵颗粒中剂量、芪茵颗粒高剂量,每组10只。空白组予以常规饲养,其余五组给予高脂乳剂持续喂养8周后随机抽取持续喂养高脂乳剂大鼠6只,观察肝脏病理组织学的改变,确定实验性NAFLD模型的造模成功;除空白组予以标准饲料喂养,并配以同等剂量的蒸馏水每日下午灌胃,其余五组均继续以高脂乳剂15ml/kg灌胃喂养。低、中、高剂量组分别给予芪茵颗粒3g/kg·d-1、6g/kg·d-1、12g/kg·d-1,易善复组给予0.069g/kg·d-1,连续12周后处死大鼠,心脏采血;测定血清瘦素(Leptin)、空腹血糖(FBS)、空腹胰岛素(FINS)。HE染色后光镜下观察肝脏组织病理学改变。结果:本实验以高脂乳剂成功复制了NAFLD大鼠模型。模型组瘦素(LP)、胰岛素抵抗指数(IRI)较空白对照组明显升高;芪茵颗粒低、中、高剂量组均可不同程度地降低NAFLD大鼠血清瘦素和胰岛素含量,改善胰岛素抵抗状态,其作用与易善复组比较,差异无统计学意义;对脂肪肝大鼠肝脏脂肪变有明显的改善作用(P<0.01)。结论:芪茵颗粒可改善NAFLD大鼠肝组织脂肪变,对NAFLD大鼠有防治作用,其机制可能与改善血清瘦素和胰岛素抵抗有关。

【Abstract】 Objectives: QiYin granular formulation has preferably curative effect on NAFLD, To investigate more probe into the mechanism of NAFLD and the therapeutic effects of NAFLD, accordingly, it can offer preferably theoretics and experimentation thereunder for clinic. Methods: 60 half male and half female Wistar rats were randomly divided into six groups: the control group, model control group, Essentiale control group,low-dose Qi Yin particles, the particles Qi Yin dose,high-dose Qi Yin particles,each Group 10;blank group to conventional farmers,and the remaining five were given high-fat emulsion continued feeding eight weeks,randomly selected sustained rats fed high-fat emulsion six observed liver histopathological changes,experimental set of non-alcoholic fatty liver Model modeling success addition to the standard blank group feeding and printed the same daily dose of distilled water at gavage,and the remaining five will continue to be high-fat emulsion to 15 ml / kg gavage feeding. Low, middle and high dose groups were given Qi Yin particles 3 g / kg·d-1·6g/ kg·d-1·12g/ kg·d-1·, Essentiale group to 0.069 g / kg·d-1,12 consecutive weeks after rats were killed, blood heart; fasting blood sugar (FBS), insulin(INS), Leptin(LP)were measured, immediately paunch rat liver, measure the weight of liver, calculate liver index, HE staining was observed under light microscope after liver histopathological changes.Result: The high-fat diet to reproduce the success of non-alcoholic fatty liver rat model. Qi Yin particles can decreased significantly experimental non-alcoholic fatty liver alleviate Insulin Resistance and Leptin .Conclusion: Qi Yin particles can be significantly reduced NAFLD rat liver steatosis, alleviate Insulin Resistance and Leptin.The prescription’s collocation is in reason, it’is a availability prescriptionof cure NAFLD.
