超声波辅助提取沙棘籽油的工艺优化研究

【摘要】 沙棘( Hippophae rhamnoides L. )是我国生态经济林建设的珍贵树种,也是著名的药食兼用果树,资源及其丰富。沙棘的药用功能价值集中反映在沙棘油上。沙棘油中富含多种生物活性物质,不仅在增强人体免疫力、抗癌、抗炎、抗辐射、治疗心血管系统疾病等方面具有良好效果,而且还具有营养脑细胞、调节植物神经紊乱等作用,因此,广泛应用于医药、保健、美容和化妆品行业。本文重点研究了沙棘籽油超声波辅助溶剂浸提工艺,通过单因素试验考察了影响提取效果的工艺参数—溶剂的种类、超声波提取时间、超声波功率、超声波频率、料液比、物料细度、提取次数等,并用响应面分析的方法进行了提取工艺优化。最终确定优化的工艺条件是:提取溶剂为石油醚,提取时间17min,超声波功率800W,料液比为1∶9 (g/mL),物料细度是40目,提取次数为2次,油脂得率高达9.27%。针对工艺因素,对所提取的沙棘籽油的四个重要质量指标—酸价、过氧化值、皂化价和碘价进行了测定分析,同时也考察了提取条件对油脂各个指标的影响。本文采用GC-MS及分光光度计法首次对超声波辅助浸提沙棘籽油的脂肪酸成分、维生素E和类胡萝卜素等功能成分进行了定量分析。测定结果为:油脂酸价6.75;皂化价174.78,碘价178.36过氧化值1.26;沙棘籽油不饱和脂肪酸达93%,必需脂肪酸含量66%,类胡萝卜素为5.53 mg/100g,维生素E 400.56mg/100g。结果表明,超声波辅助浸提沙棘籽油工艺可行,油品质量完全符合国家相关标准。

【Abstract】 Hippohae is a rare plant resource for medical and edible use in China. Sea buckthorn oil, as one of the most important aspects involved in healing properties, is rich in varied bioactive substances. It can strengthen the immune system of human beings, boost cancer-fighting ability, resist radiation and inflammatory, fight aging and prolong life . Meanwhile, it can function well on nourishing the brain cell and adjusting the plant nerve, The oil has been widely used in the industry of medicine, health care, hairdressing and cosmetic.This paper chiefly studied the ultrasonic wave‐ assisted extracting technology of the sea buckthorn seed oil. The optimum condition was determined by the single factor and the RSA response surface analysis, which were: sea buckthorn seed was used as raw materials, petroleum ether as the best extraction solvent, ultrasonic power 800W, extraction time 17min, and the liquid-solid ratio 9∶1. Under these optimum extraction condition, the extraction rate of the crude seed oil of sea buckthorn reached 9.27%.Then, we determined and analyzed the four most important index of the extracted oil---acid value, peroxide value, saponification value, and iodine value. This research indicate the composition contents of sea buckthorn seed oil by GC-MS and Spectrophotometer method. By doing this, we could know how the extracting condition affected these indexes and qualities. It is proved that sea buckthorn seed oil extracted by ultrasonic wave has higher economic value and medical health protection value. This analytical results provide scientific basis for development and utilization of the ultrasonic wave-assisted extraction technology in oil field.
