鹿茸多肽的提取分离及其促进成骨细胞增殖作用的研究

【摘要】 鹿茸系鹿科动物梅花鹿(Cervus Nippon Temminck)或马鹿(Cervus elaphus Linnaeus)的雄鹿未骨化密生绒毛的幼角。中医主要用其治疗虚劳赢瘦、腰膝酸痛、心悸耳鸣、阳痰滑精、子宫虚冷、崩漏带下等症。鹿茸中含有多种生物活性物质能够促进机体的生长发育和新陈代谢,增强机体的免疫功能,对神经系统、心血管系统有良好的调节作用,有助于恢复和保持机体健康。本文采用水溶液提取法从新鲜梅花鹿鹿茸中提取出鹿茸多肽,然后经Sephadex-G-25葡聚糖凝胶层析和制备型高效液相色谱柱分离纯化,得到一系列分子量小于800u的小肽。采用MTT法检测鹿茸多肽对人成骨肉瘤细胞(OS-732)细胞的增殖活性的影响。结果表明鹿茸多肽能明显促进OS-732细胞的增殖。改良Gomori钙-钴法显示碱性磷酸酶,结果显示鹿茸多肽促进OS-732细胞增殖的同时,碱性磷酸酶的表达也明显提高。同时采用流式细胞仪检测鹿茸多肽作用OS-732细胞后的细胞周期变化,结果表明鹿茸多肽作用后OS-732细胞其S期所占的百分比明显高于对照组,说明鹿茸多肽对OS-732细胞有非常明显的增殖促进作用。本研究为开发鹿茸治疗骨质疏松症药物的研究提供了实验依据。

【Abstract】 Deer velvet antler is the young antler which is still not ossified and full of velvet of the male deer Cervus Nippon Temminck. Velvet antler is one of the most popular natural tonics and traditional medicinal substance. It is a valuable drug in the Orient.Velvet antler contains numerous regulatory substance, chemicals and co- mponents, such as polysaccharide, phosphatide, hormone, mineral composition, polypeptide and protein.The growing antler has been used for the treatment of various diseases in oriental medicine. Deer antler as an animal source of the Chinese tradition medicine has been used in therapy of neurosis, enriching the vital energy, nursing the blood, strengthening the kidney and prolonging life for thousands of years. Many reports made it evident that velvet antlers have various effects: anti-inflammatory affects anti-whiplash activities as antistress activities and anti-aging activities, etc. A water extract of velvet antler which prepared from the velvet antler of sika deer (Cerv us Nippon Temminck). Extract has been widely used in the treatment of some osteocyte-related diseases, such as osteoporosis, osteoarthritis, and bone fracture.People with osteoporosis(OP) experience a low bone mass and a weakening of the bone structure . This can eventually result in debilitating fractures, particularly of the hip, spine and wrist. The reason of OP is general consider that the body is short of calcium and vitamin, endocrine disturbance, nutritional disturbance and lack of sports and illumination. Most people currently suffer from osteoporosis,and another’s are considered to be at risk for the disease. Women are four times more likely than men to develop the bone loss condition. Approximately 30%-50% for female and 15%-30% for male will suffer from bone fracture which initiation by osteoporosis. Osteoporosis is dividatur into 3types: primarily osteoporosis, continue osteoporosis and idio- osteoporosis.The native total peptides(VATP)were extracted from velvet antlers of sika deer ( Cerv us nippon Temminck). A high active peptide component (VAP-B)stimulating proliferation of human osteosarcoma cells was isolated from VATP. Finally, a further purified active peptide component (VAP-B1)with molecular weight distribution from 200-800 Da was obtained from VAP-B by preparative HPLC. VAP-B1 is able to enhance the average content of DNA in S phase. And also the alkali phosphatase (ALP) activity in osteosarcoma cells increased obviously with an increase in VAP-B1 dosage and treatment time. These results are consistent with cell cycle variation and ALP level as a marker of bond formation during osteocyte proliferation and differentiation, as well as maturity. This study provided a clue to explore a new peptide medicine from velvet antlers for therapy of osteoporosis.
