呼市地区奶牛乳腺炎金黄色葡萄球菌的分离鉴定及基因分型

【摘要】 金黄色葡萄球菌是引起奶牛乳腺炎的一种主要传染性病原菌。本实验从呼市地区,经CMT和体细胞检测确定患有乳房炎的77头奶牛采取223个样品进行了金黄色葡萄球菌的分离鉴定,共分离到的63株金黄色葡萄球菌。攻毒试验证实63株金黄色葡萄球菌均有较强的致病性。分离的金黄色葡萄球菌对9大类21种药物的药敏试验结果表明,本研究所分离的金黄色葡萄球菌对临床上常用的青霉素类、磺胺类药物及头孢唑啉等药物均已产生不同程度的耐药性,而氨基糖苷类药物和喹诺酮类药物则表现出较好的抑菌作用。经RAPD扩增后,纳入该研究的63株金黄色葡萄球菌可见多种带型,扩增产物的电泳条带在2~13条之间,分子量大小在50~2 200bp之间。利用SPSS11.5软件对金黄色葡萄球菌的RAPD指纹图谱进行分析表明,根据遗传距离的差异进行聚类分析,将63株金黄色葡萄球菌分成了12个主要聚类群,每个聚类群内部菌株的遗传距离相同或接近,符合此12组基因型的亲缘关系相同或相近的病源菌占(73%)。本实验通过药物敏感性试验、动物致病性筛选、随机扩增DNA的多肽性分型(RAPD)等技术,了解到呼市地区引起奶牛乳腺炎金黄色葡萄球菌不同遗传基因型的致病性及对各种抗生素的耐药情况。并与国内外其它地区的基因型进行比较与分析,为进一步研究耐药性基因及研发对本地区预防效果较好的DNA疫苗奠定基础。

【Abstract】 The staphylococcus is a kind of main contagion cause germ which causes milk cow mastitis. Experiment to shout with specially Huhhot area, through CMT and body cell examination assurance of 223 samples from 77 mastitis cow .Carried out the authenticate of separation staphylococcus ,cure evaluate and the medicine sensitivity experiment. Also studied the disease staphylococcu to carry on to RAPD random to 63 stubs separated.The conteracting toxic substances experiment shows 63 kind stubs is pathogenicity staphylococcus.The experiment use 9 major type 21 kinds of medicine which is clinical treatment on pathogenic staphylococcus. penicillin, the sulfasulfonamide medicine and cephazoline. the medicine all have already produced different degree drug resistance ,but aminoglycosides and Du-6859a have obviously bacteriostasis effect.After RAPD found that 63 strain staphylococcus has muti-band froms, expanding between 2~13, molecular weight is between the 50~2200 bps.Use SPSS11.5 analyze the staphylococcus band , according to genetic distance cluster analysis the result show 12 mainly gather types ,and 73% genotypic hasgenetic relationship.Therefore, this experiment passes drug sensitivity experiment, and RAPD technique, studied huhhot area staphylococcus drug resistance and different genetic gene ,and comparison with domestic and international the genotype.Foundation for research of drug resistance gene and better DNA vaccine.
