猪场污水中脱氮微生物的分离与鉴定的研究

【摘要】 水是宝贵的自然环境资源,是人类生存和发展的源泉。氮素是水体中污染的一类重要污染物。不仅可用引起水体富营养化,还可直接影响动物及人体的健康。猪场污水是集约化养猪场饲养过程中由生猪所排放出的液体代谢产物和冲洗固体粪便等生活残渣所产生的高浓度有机污水。随着畜牧业的发展,猪场污水也是也是氮素的一个主要来源。水污染越来越严重,如何有效处理和处置畜牧养殖业的污水,使其达到排放标准,达到回收利用,减少环境污染的等问题正摆在我们面前。脱氮是污水处理过程中一个重要的环节。生物脱氮是目前污水脱氮处理中最常见的方法。为了得到脱氮能力较强的微生物,本试验通过极限稀释法以及平板划线法,共筛选到了34株微生物,通过对这34株微生物的复筛试验,得到一株脱氮效果最好的菌株B2,在高浓度的人工废水中,48h内氨氮去除了为79.94%。经鉴定为不动杆菌属。对菌株B2进行了相关性质的研究,确定了最佳脱氮条件:最佳温度是35℃、最佳C/N为19.3,最佳为pH=8.1,对小分子碳源有较强的依赖性。对B2进行了实际污水脱氮效果的评价,由于实际污水成分复杂,去除率为75.77%。在真正污水处理过程中,菌株要经过驯化的过程。

【Abstract】 Water is the valuable natural resources, and it is the sources of human existence and development. Nitrogen is one of major pollutants of water pollution, and can cause water eutrophication and can directly influence the health of animal and human. Pig farm sewage is the high density organic sewage that is the result of liquid supersession discharged out by live pig and solid excrement and urine in intensify pig farm raised course. With the development of animal husbandry, the sewage of pig farm is a plain main source of nitrogen too. The water pollution is more and more serious, how to deal with and handle the sewages of the animal husbandry feeding effectively, how to make it reach to the discharge standard and recycle and reduce the environmental pollution, those questions are being put in front of us. The denitrification is an important link in the course of sewage disposal. The biological denitrification is the most common method in the sewage denitrification at present.In order to receive the microorganism of the denitrification with stronger ability, 34 strains of microorganism which could heterotrophic nitrify were isolated. Through to the second sift in 34 strains, we found strain B2 could remove nitrogen most rapidly in those 34 strains. In the artificial waste water of the high density, it is 79.94%that ammonia nitrogen is removed in 48h.Strain B2 was identified as Acinetobacter genus.Carrying on the research of relevant properties to B2, we had confirmed the best denitrification condition. The most optimal nitrogen removal condition is 35℃, C/N=19.31, pH=8.10, There is a stronger dependence to the carbon source of small molecule for B2.Inoculating B2 with the sewage of pig farm, nitrogen removal ability of B2 was measured in the real sewage. Because of the real sewage complicated composition, it is 79.94%that ammonia nitrogen is removed. So in real sewage disposal, the course that the bacterial strain should be tamed.
