干扰素-τ基因多态性和生殖激素与受胎率的相关性研究

【摘要】 本研究采用PCR产物直接测序技术,首次对109个中国荷斯坦奶牛个体的牛干扰素-τ(bIFN-τ)基因编码区(CDS)进行测序,并结合繁殖生产数据,分析不同位点的基因型和情期受胎率、不同胎次与血清中孕酮和雌二醇浓度、情期受胎率与血清中孕酮和雌二醇浓度的相关性,分析结果如下:1.SNP分析表明,在中国荷斯坦奶牛的IFN-τ基因CDS区265bp和301bp存在两个变异位点,分别为T→C(记为位点一)突变和C→T(记为位点二)突变,并且杂合子基因型在两个变异位点上均为优势基因型。2.变异位点一、位点二杂合度分别为0.7430和0.8350;多态信息含量分别为0.3700和0.3680,两个位点均为中度多态,且两者在实验材料样本中的分布极显著地偏离了Hardy-Weinberg平衡定律(P<0.01)。3.最小二乘方差分析表明,不同位点的基因型与中国荷斯坦奶牛受胎率之间差异均不显著(P>0.05)。4.最小二乘方差分析表明:中国荷斯坦奶牛发情期血清中孕酮水平0胎次与4胎次,0胎次与5胎次,1胎次和5胎次,2胎次和5胎次之间为显著差异(P<0.05);1胎次和4胎次,2胎次和4胎次,3胎次和4胎次,4胎次和6胎次,5胎次和6胎次,6胎次和7胎次之间为极显著差异(P<0.01)。中国荷斯坦奶牛发情期血清中雌激素水平0胎次和2胎次,1胎次和2胎次之间均存在着显著差异(P<0.05)。情期受胎率与血清中孕酮和雌二醇浓度之间差异均不显著(P>0.05)。

【Abstract】 This experiment did research on the sequence of Chinese Hostein interferon- taugene CDS firstly by using the PCR production Direct sequencing technology. Basedon the detection of registered data, the effects of different genotypes with conceptionrate、the different deuto foetus with progestone level and estradiol level, pregnancycondition with progestone level and estradiol level were analyzed.1.There are two mutation locuses in interferon- tau gene CDS of the Chinese Hostein,they are T→C mutation and C→T mutation. besides that zygotic genotypes aresuperiority genotypes in different mutation.2.Heterozygosis degree of the different mutation locuses is 0.7430 and 0.8350; Thepolymorphisms information content is 0.3700 and 0.3680, so mutation locus one andmutation locus two are midrange polymorphisms. The results of x~2 fitness test of locusone and locus two indicated that the Chinese Hostein were high significantly biasedfrom Hardy-Weinberg equilibrium (P<0.01).3. least squares analysis showed that the different locus genotype is quiet with theChinese Hostein conception rate (P>0.05).4. Least squares analysis showed that level progesterone of the Chinese Hostein issignificant devitation (P<0.05) in 0 foetus and 4 foetus, 0 foetus and 5 foetus, 1foetus and 5 foetus and 2 foetus and 5 foetus.Least squares analysis showed that levelprogesterone of the Chinese Hostein is extreme significant devitation (P<0.01) in1 foetus and 4 foetus, 2 foetus and 4 foetus, 3 foetus and 4 foetus, 4 foetus and 6foetus, 5 foetus and 6 foetus and 6 foetus and 7 foetus. Least squares analysis showedthat level estradiole of the Chinese Hostein is significant devitation (P<0.05) in 0foetus and 2 foetusang and 1 foetus and 2 foetus. Least squares analysis showed thatthe different pregnancy condition is quiet with level progesterone and estradiole in theChinese Hostein (P>0.05).
