凉水国家自然保护区天然红松种群遗传多样性在时间尺度上变化的cpSSR分析

【摘要】 本文应用叶绿体微卫星(cpSSR)技术对凉水国家自然保护区天然红松(Pinus koraiensis)种群的遗传多样性在时间尺度上的变化进行了遗传分析。共选择121个树龄在1581-1900年间的红松个体,连续分为6个龄级。从6对叶绿体SSR引物筛选出两对引物,共检测到7个位点,其中多态位点5个,多态位点比率为71.43%。利用POPGENE分析软件计算了红松各龄级内的遗传多样性指数,分析了遗传多样性在红松各龄级间的变化。由Nei指数估算出红松龄级内基因多样性的平均值为0.1985,龄级间基因多样性的平均值为0.0164,种群基因多样性平均值为0.2149,可以算出,龄级内的遗传变异占92.37%,而龄级间只有7.63%,表明红松龄级间遗传多样性分化不明显,遗传多样性主要存在于龄级内。由红松各龄级的Shannon指数和Nei指数的走势可以看出该保护区内的红松遗传多样性在这320年间没有发生太大的波动,基本呈现稳定的趋势。说明该时期的天然红松种群未受到人类活动的破坏,属于自然演替过程,遗传多样性基本稳定。

【Abstract】 With cpSSR method, the variation of genetic diversity in temporal dimension of natural population of Pinus koraiensis in Liangshui national nature reserve was studied in this paper. Total 121 P.Koraiensis samples aged from 101 to 420 years were grouped into 6 age classes. 5 polymorphic loci were obtained from 2 pairs of SSR primers and the percentage of SSR polymorphic loci was 71.43%.With the POPGENE analysis software, the genetic diversity index was calculated and the variation of genetic diversity was analyzed in every age class level. By Nei’s index estimated, the average level of genetic diversity within age classes is 0.1985, the average level of genetic diversity among age classes is 0.0164, and the average level of genetic diversity in the population is 0.2149. It can be calculated that the genetic differentiation within age classes accounted for 92.37%, and the genetic differentiation among age classes only accounted for 7.63%.This result showed that the differentiation of genetic diversity is not obvious in age class level, and genetic diversity of P.koraiensis primarily existed within age class. The trend of Shannon and Nei’s index among age classes showed that the genetic diversity of P.koraiensis in Liangshui National Nature Reserve didn’t fluctuate very much during 320 years. During that period, the natural population of P.Koraiensis has not been damaged by human activities, is a natural succession process, and has the basic stability of the genetic diversity.
