扁平苔藓中caspase-9、Apaf-1的表达及其与细胞凋亡的关系

【摘要】 目的:研究扁平苔藓(lichen planus LP)皮损中细胞凋亡的情况,探讨caspase-9、Apaf-1在扁平苔藓皮损中的表达和意义,分析二者的表达与扁平苔藓中细胞凋亡的关系。方法:应用原位末端标记技术(TUNEL法)及免疫组化的方法检测30例LP皮损和30例正常皮肤组织中的细胞凋亡的情况及caspase-9和Apaf-1蛋白的表达部位和表达强弱;采用RT-PCR的方法检测caspase-9mRNA在10例LP皮损和8例正常皮肤组织中的表达情况。结果:LP中表皮层角质形成细胞的凋亡指数(AI)75.67±9.4,明显高于正常对照组38.15±9.6(P<0.01);LP中Caspase-9和Apaf-1的阳性单位分别为14.45±2.72和11.93±3.14,均明显高于正常对照组(P<0.01);而且LP中caspase-9mRNA的表达量2.304±0.417与正常对照组1.605±0.381比较差别有统计学意义(P<0.05)。LP中caspase-9、Apaf-1的阳性表达强度均与其自身的角质形成细胞的AJ呈正相关r1=0.965 r2=0.939(P<0.01)。结论:LP中角质形成细胞凋亡亢进;线粒体凋亡途径的凋亡因子caspase-9、Apaf-1与角质形成细胞的凋亡关系密切;二者介导的线粒体凋亡途径可能参与了LP的发病过程。

【Abstract】 Objiect: To study the apoptosis of the keratinocytes in lichen planus and examine the expression and distribution of caspase-9、Apaf-1 in lichen planus. Analysis the relationship between the expression of them and the events of cell apoptosis in lichen planus.Methods: TUNEL and Immunohistochemical technique were employed to study apoptosis of keratinocytes and the expression of caspase-9、Apaf-1 in 30 linchen planus and 30 normal skin specimens.Caspase-9 mRNA were observed by RT-PCR in 10 linchen planus and 8 mormal skin.Results:The keratinocytes apoptosis index (AI) 75.67±9.4 in lichen planus were increased than its 38.15±9.6 in normal contral(P<0.01).Compared with the normal control group,positive units(PU) of caspase-9 and Apaf-1 was 14.45±2.72 and 11.93±3.14 which were upregulated in keratinocyte in lichen planus (P<0.01);Caspase-9mRNA 2.304±0.417 were also increased in LP than its in nomal skin(P<0.05);Both caspase-9 and Apaf-1 were direct correlation with their own keratinocytes apoptosis index r1=0.965 r2=0.939 (P<0.01) .Conclusion: The apoptosis of keratinocytes were significantly accelerated in lichen planus; Caspase-9 and Apaf-1 might be participated in the apoptosis of kerationcytes in LP cases; Mitochondria apoptosis pathway may contribute to the formation and progression of lichen planus.
