灌胃绞股蓝提取液对自然衰老小鼠皮肤组织中H2O2含量、GSH-PX活性影响的实验研究

【摘要】 目的:本项研究旨在通过给药组动物灌胃中药绞股蓝提取液,测定皮肤组织中过氧化氢(H2O2)的含量、谷胱甘肽过氧化物酶(GSH-PX)的活性,及皮肤组织超微结构的变化,来检测绞股蓝提取液在减轻小鼠皮肤自然老化方面的作用,并进一步认识和论证中医补虚扶正抗衰老的临床意义和现代医学中导致皮肤老化的自由基损伤学说。方法:取健康昆明种雌性3月龄小鼠10只作为青年对照组(A);12月龄自然衰老小鼠40只,随机分为4组:老年灌胃空白对照组(B)、老年灌胃阳性对照组(C)、老年灌胃绞股蓝组(D)、老年(灌胃+外用)绞股蓝组(E)。其中A组、B组灌胃生理盐水(0.8ml/t),C组灌胃维生素E(0.2ml/t),D组、E组分别灌胃绞股蓝提取液(0.8ml/t),并且E组外涂加入赋型剂的绞股蓝霜(0.5g/t),涂抹面积为(3×3cm2),其浓度为8%。每日二次,共计30d。采用病理组织切片制片技术,HE染色,以100倍光镜观察真皮结构改变,并以生化分析方法测定H2O2含量及GSH-PX活性。结果:与青年对照组比较,其它四个老年组小鼠皮肤组织中H2O2含量显著增加(分别是P<0.01,P<0.05,P<0.01,P<0.05),GSH-PX活性明显降低(分别是P<0.01,P<0.01,P<0.01,P<0.01);四个老年组小鼠皮肤组织病理切片呈现自然老化状态;与老年灌胃空白对照组比较,老年灌胃阳性对照组、老年灌胃绞股蓝组及老年(灌胃+外用)绞股蓝组均可降低H2O2含量(分别是P<0.01,P<0.01,P<0.01),提高GSH-PX活性(分别是P<0.01,P<0.05,P<0.01);与单纯灌胃绞股蓝的D组相比,同时配合以外用的给药方式的E组对降低H2O2含量(P<0.05)及提高GSH-PX活性的作用较强(P<0.05)。结论:中药绞股蓝提取液可通过清除自由基,提高机体抗氧化能力,减轻自然衰老小鼠皮肤组织氧化损伤,并可通过降低H2O2的含量和提高GSH-PX的活性,从而延缓皮肤组织的衰老。灌胃配以外用绞股蓝提取液该种给药方式能更好的起到延缓自然衰老小鼠皮肤的损害。前言几个世纪以来衰老一直是科学家感兴趣的课题,科学家对衰老过程的兴趣不断增加,特别是近20年来老年人群在总人口中所占的百分数明显增加。近年有学者提出,衰老是伴随着年龄增加(简称随龄)机体发生的功能性和器质性衰退的进行性改变的积累。皮肤衰老是机体衰老的一个重要组成部分,皮肤外露,在机体衰老过程中容易显露,易于观察,已成为研究衰老的重要靶器官。皮肤衰老不仅影响美容,且与许多皮肤病如脂溢性角化、日光性角化、鳞状细胞癌的发生也有着病因学的关系。因此预防和延缓皮肤衰老已成为生命科学和医学美容的研究热点。在对衰老机理的追探中harman于1956年提出的自由基学说受到了普遍的认同和关注,其基本观点是,衰老是由自由基引起的组织随机毒害的结果。在正常的生理情况下,体内有少量的氧自由基产生,它们参与正常代谢。同时,体内也存在着清除自由基、抑制自由基反应的体系,使得过多的自由基被清除,或使自由基产生减少。清除抑制自由基反应的体系主要有酶(超氧化物歧化酶SOD、过氧化氢酶CAT、谷胱甘肽过氧化物GSH-PX)

【Abstract】 Objective: The experiment studied giving traditional chinesemedicine GP to the animal to determine whose content of hydrogenperoxide (H2O2)of the skin organization, the change of Glutathioneperoxidase (GSH-PX) and the skin ultra structure, to investigate GPeffect on the respect of small rat skin aging, delay a senilitythereby, and lead to skin aging free radical loss theory in clinicalsignificance and modern medicine old and feeble furter knowing andexpounding and proving doctor of traditional chinese medicine to addsupplemental false strengthening the body resistance resisted.Methods: 10 Healthy KM female rats age in 3 months contrasting a groupas youth (A);40 old in natural of KM female rats age in 12 monthswere divided into 4 groups randomly and gavagad respectively, thatis old age oral blank group (B),old age oral gender group (C),oldage irrigates oral GP group (D),old age oral GP and external group(E).A group and B group oral saline (0.8ml/t),C group oral VitaminE (0.2ml/t),D group and E group oral the GP (0.8ml/t),and E groupexternal the GP (0.5g/t),daub area for (3×3cm2), whose thicknessis 8%.Two times per day, altogether 30 day. Adopt pathologicalmechanism to organize the section film production technology, HEdyeing, the structure observing a dermis changes, and analysismethod determines with biochemistry H2O2 contents and GSH-PX acti-vity with 100 glossy mirrors.Results: Comparing with youth collation group, H2O2 contents arenotable in other four old age rats of groups skin organizationincreasing (respectively P<0.01, P<0.05, P<0.01, P<0.05), thatGSH-PX activity reduces (respectively P<0.01, P<0.01, P<0.01, P<0.01);Pathological mechanism section of organization displaysthe state aging naturally; Comparing with oral blank group, old ageoral gender group, old age irrigates oral GP group and old age oralGP external group is equal but reducing H2O2 contents (respectivelyP<0.01, P<0.01, P<0.01, P<0.01), improving GSH-PX activity(respectively P<0.01, P<0.05, P<0.01);And irrigate that the oldage irrigates oral GP group D being compared with each other, usesbesides fitting at the same time to reduce contents H2O2 (P<0.05)for medicine way E group and improves stronger active GSH-PX effect(P<0.05) exclusively.Conclusion: The GP the oxidation resistance picking up liquid butraising airframe by eliminating a free radical, effects small rat skin organization and feeble oxide loss,reducing the H2O2 contentsand the activity improving GSH-PX, delaying the senility that skinorganizes thereby. Oral the GP and external can get up more well todelaying damage, whose mechanism at ease old and feeble to smallrat skin possibility for medicine way is that way can raise bodyinner medicine since inside and outside is the medicine with begiven to concurrently thickness more beneficial to airframe sorp-tion make use of.
