独一味茎叶活性成分研究

【摘要】 独一味来源于唇形科植物独一味Lamiophlomisrotata(Benth.)Kudo的根及根茎或全草,为药典收载品种。味苦,微寒,有小毒,温,入口久则有麻痹感具有止血、镇痛消肿、活血化瘀、补髓、行气、续筋接骨等功效。研究结果表明环烯醚萜成分是独一味的有效部位之一,具有较强的镇痛止血作用。2005版药典对独一味质量控制方法是通过性状、显微及黄酮类成分定量研究,所以寻找药理活性成分并建立有效的含量测定方法,对控制对一味药材及制剂的质量具有重要意义。对独一味茎叶醇提取物通过聚酰胺柱后的两个提取部位进行了小鼠镇痛和止血试验研究。确定水洗脱部分镇痛和止血效果优于醇洗脱部分。继而对水洗脱部分通过AB-8大孔吸附树脂柱后的五个提取部位进行了小鼠镇痛和止血试验研究。确定30%醇洗部分为镇痛和止血的主要有效部位。对独一味茎叶的有效部位——环烯醚萜类成分进行了系统的化学成分研究。采用硅胶柱层析,TLC,凝胶柱层析等分离手段分离得到了7个单体化合物,并通过理化性质及波谱解析分别鉴定为:β-谷甾醇、乙酰基山栀苷甲酯、6-O-L酰山栀苷甲酯、8-O-乙酰山栀苷甲酯、脂麻苷、7-去羟基马钱苷、7-去羟基马钱酸。其中7-去羟基马钱酸为首次从独一味中分离得到。分别采用小鼠醋酸扭体法、断尾法、耳肿胀法,对从独一味茎叶中首次分离得到的环烯醚萜类化合物7-去羟基马钱酸进行活性试验。确定该化合物为独一味镇痛、止血和抗炎的有效成分之一。初步确定环烯醚萜类化合物为独一味镇痛止血作用的物质基础。确定独一味药材最佳提取工艺为60%乙醇浸润药材36小时,以15倍体积的60%乙醇渗漉提取。富集工艺为将60%乙醇提取物上聚酰胺柱,水洗脱部分上XDA-7大孔吸附树脂,收集30%醇洗脱部分。

【Abstract】 The plant Lamiophlomisrotata (Benth.)Kudois used in abirritation and hemostasia. Iridoids is Lamiophlomisrotata(Benth.)Kudo. Yhe application of reactive component like Iridoids in Lamiophlomisrotata(Benth.)Kudoinclud the pharmacdogical action on abirritation and hemostasia. Moreover, the pharmacopoeias of 2005 edition lacks the quantitative investigation, So we studies its steroidal saponins ingredient and seek the monomer active constituent. It is significant to find and establish the effective content determination method for quality control of the preparation and crude drugs.Screening test of Analgesic and hemostatic portion. The 70%ethanol extract in stem and leaves of Lamiophlomis rotata (Benth.)Kudowas isolated by polyamide into two portion, the portion eluted by water has the better Analgesic and hemostatic effect.So using AB-8 macroporous resin,we purified the above portion and obtained five ethanolic elutions, the 30% ethanolic elution show strong Analgesic and hemostatic action.We studied on Iridoids which is the active part of Lamiophlomisrotata (Benth.) Kudo. The method of silica column chromatography, thin layer chromatography and gel column chromatography was used to isolate the chemical constinents. Their structures were identified by physicochemical properties and spectra analysis (1H-NMR and 13C-NMR). Eight compounds were isolated and their structures were identified as -sitosterol, 8-0-acetyl-shanzhiside, methyl ester, shanzhiside, 6-0-acetyl- shanzhiside methyl ester, methyl ester, sesamoside, dehydropentste-moside, 7- deoxyloganic acid is known first finded compound in Lamiophlomisrotata (Benth.).Pharmacological screening test of compound 6. Compound 6 was evaluated by the mouse stretching-body method, tail-cutting method and auricular-swelling method, compound 7-deoxyloganic acid can be affirmed as the active compound of hamiophlomis rotata (Benth.) Kudo.Studies on extraction and purification process, the best extraction was established by orthogonal test. solvent: 60% ethanol, soak:36 hours, elution volume:f5 times of materials .Pufification process:the ethanolic extract should be isolated by polyamide. Then isolate the water elution with XDA-7 macroporous resin and collect the 20% ethanolic extract.
