解毒养肝汤治疗药物性肝损害的实验研究

【摘要】 目的：解毒养肝汤治疗药物性肝损害的疗效探讨。方法：将3月龄昆明种小鼠分为正常对照组、模型对照组、解毒养肝汤高、中、低剂量组及护肝片组，每组10只。正常对照组给予普通饲料喂养，正常饮水，不施加其它人为影响因素。除正常对照组外，其余各组予腹腔注射乙酰氨基酚混悬液(150mg·kg^-1)，16h后，低、中、高剂量组分别按3.33g／（kg·d）、6.66 g／（kg·d）、10.0g／（kg·d）的给药量予解毒养肝汤灌胃，阳性对照药组给予护肝片（22.75mg／kg体重）水溶液灌胃，模型对照组和正常对照组分别喂饲同体积的蒸馏水。每日1次，共2周。最后一次灌胃后，各组动物均禁食12小时，腹主动脉采血。通过对药物性肝损害小鼠ALT、AST、ALP、GGT、TBIL、ALB、A／G的检测，探讨解毒养肝汤的保肝降酶、防治肝损伤的作用。结论：解毒养肝汤各剂量组均能降低对乙酰氨基酚混悬液所致的小鼠ALT、AST、ALP、GGT、TBIL的升高，提高对乙酰氨基酚混悬液所致的小鼠的ALB、A／G的降低，作用强度与浓度有关，经统计学处理以高剂量组疗效最为显著，在小鼠生化指标疗效上，解毒养肝汤优于护肝片组。说明解毒养肝汤有很好的治疗药物性肝损害的作用。更多还原

【Abstract】 Objectives: To explore the efficiency of Jieduyanggan Soup in treating Drug-induced liver injury. Methods : Sixty healthy three-month-old Kunming mice were divided into six groups of ten as following:normal control group, model control group, high-dosage treatment group,middle-dosage treatment group,low-dosage treatment group, Huganpian positive control group. Expect group one, the other five groups were injected Paracetamol Suspension (150mg.kg^-1) in the abdominal cavity of the mice. Sixteen hours later, mice of group three, group four and group five were forced to take the Jieduyanggan Soup directly intostomach respectively according to the dosage of 3.33g/（kg·d）,6.66g/（kg·d）, 10.0g/（kg·d）. Mice of group six were forced to take Huganpian aqueous solution （22.75mg/kg）. Mice of group one and group two were fed the same volume of distilled water. Keeping these actions once per day for two weeks. After the last time of giving medicine, mice of each group were forbidden to eat for twelve hours. Then took the blood sample through the mice’s abdominal aorta. Through the examining on the Drug-induced liver injury mice’s ALT, AST, ALP, GGT, TBIL, ALB, A/G to explore the function of Jieduyanggan Soup on protecting liver, lowering GPT and preventing liver damage. Conclusion: Each dosage of Jieduyanggan Soup can lower the rising of the mice’s ALT, AST, ALP, GGT, TBIL caused by Paracetamot Suspension,and can rise the mice’s falling of ALB, A/G caused by Paracetarnol Suspension.The curing intensity id related to concentration and the statistics shows that the high-dosage treatment group has better curative than the others. All of above shows that Jieduyanggan Soup has an good effect on curing Drug-induced liver injury.更多还原
