九节菖蒲化学成分的研究

【摘要】 中药九节菖蒲为毛茛科(Ranunculaceae)植物阿尔泰银莲花(Anemone altaica Fisch.exe C.A.Mey.)的干燥根茎,具有芳香开窍、醒神、散湿浊、开胃、解毒杀虫等功效,该药在临床上应用广泛,但由于九节菖蒲与现行药典所载石菖蒲(Acovus gramineus Soland)有某些相似的功能和主治,所以常常被混淆。经前人研究表明该九节菖蒲与石菖蒲在原植物形态、显微组织结构、化学成分等方面存在明显差异,为进一步研究九节菖蒲中有效成分,规范其质量标准,作者对其进行了相关的成分研究。本人先利用基本化学方法对九节菖蒲中的化学成分进行了系统预实验,对其中可能含有的成分类型进行了检识和分析,之后利用各种色谱手段,包括硅胶柱色谱、制备薄层、反相柱色谱等方法从中药九节菖蒲中分离得到五个化合物,并进一步通过波谱分析(1H-NMR,13℃-NMR,ESI-MS)和化学方法鉴定了五个化合物的结构,分别是:β-谷甾醇(Ⅰ)、琥珀酸(Ⅱ)、白头翁素(Ⅲ)、琥珀酸酐(Ⅳ)、齐墩果酸(Ⅴ)。其中Ⅳ、Ⅴ为首次从九节菖蒲中分离得到。另外通过对九节菖蒲挥发性成分和脂肪油成分的GC-MS分析鉴定了25种挥发性成分和17种脂肪油成分,其中有23种挥发油和17种脂肪油是在此植物中首次得到。本项研究为九节菖蒲单体有效成分的药效学研究奠定了化学基础,为提高九节菖蒲质量标准并探讨其药效作用成分奠定了基础,且为从化学成分角度区分九节菖蒲与石菖蒲提供了科学依据。

【Abstract】 Jiujie Changpu belongs to genus Ranunculaceae plants of Anemone altaica Fisch.exe C.A.Mey.. It can remove obstruction, soothe the nerves, eliminate dampness, stimulate the appetite, detoxicate and expell worms. Because its function is similar to Acorus gramineus Soland, these two traditional Chinese medicine are always confused. But it is indicated by experiment that they are very different in plant configuration, microstructure and chemical ingredients. In order to searching more constituents and establishing integrated quality standard of Anemone altaica Fisch, the author carried out the following studies.The author have a preliminary systematic test first by basic chemical methods, detect and analysis all kinds of constituents in Anemone altaica Fisch. Then extract and separate five compounds by the way of chromatography, including silica gel column and preparative thin layer chromatography and reversed-phase column chromatography etc. These five compounds structural elucidation was performed by spectroscopic analyses, primarily NMR, ESI-MS. Five compounds are established asβ-sitosterol (Ⅰ), Succinic acid (Ⅱ), Anemonin (Ⅲ), Succinic anhydride (Ⅳ), Oleanolic acid (Ⅴ). CompoundⅣandⅤare isolated from the plant for the first time. At the same time, we get 25 flavonnoids and 17 fatty acid by GC-MS,of which 23 flavonnoids and 17 fatty acid are isolated from the plant for the first time.The study lays chemical foundation for the medical curative effect research of the monomeric constituent of Anemone altaica Fisch.exe C.A.Mey., the study establish the foundation for the quality standard improvement and the medical curative effect constituents exploration of Anemone altaica Fisch.exe C.A.Mey.. In addition, the study gives out the scientific basis to differentiating Anemone altaica Fisch and Acorus gramineus Soland.
