葛根素的提取及纯化新工艺研究

【摘要】 葛根是一种既有丰富营养,又有独特药理作用的药食两用资源,用途广泛。作为葛根的主要活性成分,葛根素以其具有改善心脑血管循环、降低心肌耗氧量,降低血糖,防止高血压及动脉硬化,抗肝脏毒,抗炎,祛痰,解热,提高肌体免疫力,抗菌,抗病毒等多种药理作用,还具有提高视力等功能,日益受到广泛关注,其高纯度产品在国际市场上倍受青睐。本文对葛根素的提取纯化工艺进行了系统的研究,采用了冷浸—热提法和结晶—重结晶法分别进行葛根素的提取和纯化,着重研究了葛根素提取条件及结晶的条件。筛选出了较优的工艺参数,从而为葛根素的工业化生产奠定了良好的基础。研究结果表明:无论是提取工艺还是纯化工艺,与其它一些提取方法和纯化方法相比均有自己的独特之处,而且保证了相对较高的提取得率和产品纯度。本方法的创新点在于:提取过程中,采用了热的正丁醇溶液作为提取剂,可以在保证较高的提取得率的同时降低提取液中杂质的含量;纯化过程中,首先引入了蒸发结晶,即在传统的结晶前将提取液旋蒸浓缩到一定体积后冷却结晶,过滤可以除去大部分的大豆苷元和大豆苷及部分杂质,提高了粗浸膏的纯度,有利于葛根素结晶的进行;其次是在两次结晶过程之间引入酸水解过程,此过程可以将初结晶晶体中的部分葛根素衍生物水解成葛根素,既提高了葛根素的得率,又提高了产品的纯度。

【Abstract】 Pueraria is a kind of resource for drug and food since it can provide people with a lot of nutriments and particular medicative effects. It has many uses. As the main active component of pueraria, puerarin is paid more attention increasingly for it can improve the blood vessel circulation of heart and brain, reduce the oxygen consumptioncapacity of cardiac muscle, reduce the blood sugar, prevent the hypertension and artery teriosclerosis, the liver poison, inflammation, removing the phlegm, allay a fever, enhances the organism immunity, antibacterial, antivirus and so on many kinds of pharmacological action, and it also has the function of enhancing vision . The pure product is very popular in international market.In this paper, the extraction and separation technics of puerarin was studied by the numbers. Adopting quench-hot extract and rime-afresh rime progress extraction and separation respectively on puerarin. Emphatically studied the condition of the extraction and separation of puerarin. The optimal operation parameters were choosed and consequently establish the favorable base for the industrialized production of puerarin. The experimental results indicate that compared with some other methods of extraction and separation, both the extraction technics and the separation technics have its inimitable traits, furthermore it still ensures relative upper extraction ratio and purity of product. The innovating point of this method lies: In the extracting process, used the heated n-butyl alcohol as the extracting solvent, it can ensure the upper extraction ratio and reduce the impurity content in the extraction solution. In the separating process, firstly importing evaporation rime, namely rotation evaporation concentrate the extraction solution to some volume and cooling rime before the traditional rime, it can remove the most daidzein, daidzin and some impurity by filtration and improve the purity of the wide extractive, and be useful for the pueraria rime. Secondly, importing hydrolyzing process between the rime and rime-afresh, it can hydrolyze the ramification of pueraria into pueraria, it can not only improve the ratio of pueraria, but improve the purity of product.
