沙棘黄酮提取工艺及抗氧化性能的研究

【摘要】 本文以沙棘黄酮类化合物为研究对象,利用超声波法提取沙棘总黄酮,并对超声波法与常规法提取沙棘黄酮的得率进行比较,得到沙棘黄酮提取的最佳工艺条件。同时利用树脂、聚酰胺柱层析及高效液相色谱的分离纯化技术分离出沙棘黄酮单体,通过抗氧化实验得到抗氧化能力最强的沙棘黄酮单体。在超声波提取沙棘总黄酮最佳工艺条件建立过程中,选择了超声波提取时间、乙醇浓度、提取温度、物料比、超声波功率等因素进行梯度实验。最佳提取条件65%的乙醇溶液作提取剂,以1:20(g/mL)的料液比于78℃的条件下提取时间40min,超声波功率为60%,在此条件下沙棘果渣提取总黄酮得率为0.4829%。其提取时间短、能耗低可降低生产成本,提高经济效益。本实验还建立了聚酰胺柱层析分离沙棘黄酮的各项工艺参数,并得到三种沙棘黄酮单体化合物,其中第三个主峰在其的浓度为100mg/L时可以清除邻苯三酚自氧化反应产生的超氧阴离子自由基,清除率为32.2%。为开发和利用该资源提供了有益的参考。

【Abstract】 Flavanones in Sea-buckthorn was studied in this paper. Total flavones was extracted with ultrasonic wave method. It compared the productivity of flavones which were extracted with Conventional method to ultrasonic wave method, and demonstrated the best condition of extracting flavones in Sea-buckthorn. The monomer compounds were gained by the purification technology which used Resin, Polyamide column chromatography and high performance liquid chromatogramphy. The monomer compounds which have the strongest antioxidant capacity also was gained by the experiment of antioxidant.The experiment chose the ultrasonic wave to withdraw the time, the ethyl alcohol density, withdraws factor and so on temperature, material ratio, ultrasonic wave power carries on the gradient experiment, Discovers supersonic method withdraws the sea-buckthorn marc total flavanone the optimum condition. The best withdraws condition is 65% ethyl alcohol solution does with the quality score withdraws the medicinal preparation, by 1:20 (g/mL) the material fluid compares under 78℃conditions withdraws time 40min, the ultrasonic wave power is 60%, the sea-buckthorn marc withdraws the total flavanone under this condition rate is 0.4829%.But the convention returns to the conversion of sixteenths to decimal best to withdraw under the condition in it to have to withdraw for 3 hours, the total flavanone rate reaches as high as 0.6911%.The result indicated the ultrasonic wave law withdraws in the sea-buckthorn fruit the total flavanone rate to be inferior to the convention returns to the conversion of sixteenths to decimal to withdraw in the sea-buckthorn fruit the total flavanone rate high, but the ultrasonic wave law has does not destroy the effective component the characteristic, it withdraws the time short, the energy consumption is low may reduce the production cost, enhances the economic efficiency. The adsorption chromatography column of polyamide was also carefully studied in this article, and gained three kinds of flavones. The third crest of the wave can scavenge the superoxide anions that were produced from the self-oxidation reaction of Pyrogallol and when the concentration of the third compound was 100mg/L, the eliminating ratio was 32.2%. From the experiments, We have known the best extracted technology of seabucktborn flavones, and. it was develops and uses this resources to provide the beneficial reference.
