不同品种浙贝母的药学与药效学比较研究

【摘要】 浙贝母为百合科多年生草本浙贝母Fritillaria thunbergii Miq.的干燥鳞茎。味苦,性寒;归肺、心经。具有清热散结,化痰止咳之功效。贝母素甲,贝母素乙是中药浙贝母中的主要成分,具有多方面的药理作用。本文共进行了以下几方面研究:1对浙贝母药材性状鉴别和显微鉴别,同时对贝母素甲、贝母素乙进行定性薄层鉴别,结果均符合2005版药典规定。2采用正交实验设计考察提取溶剂、提取时间、氨水用量、氨水浸润时间确定浙贝母的最佳提取工艺,结果为:以2倍量氨水浸润15min,加入2倍量氯仿,置索氏提取器中回流提取4h。3 HPLC法测贝母素甲、贝母素乙的含量,根据甲乙素含量选择四月中上旬于鄞县、樟村、金华采收浙贝母作为药效学比较药材来源。4通过浓氨水喷雾引咳,测小鼠的咳嗽潜伏期、咳嗽次数,考察四月中上旬于鄞县(水洗;蛤粉)、樟村、金华采收的浙贝母的止咳疗效,结果为:低剂量组间潜伏期无差异,中剂量组潜伏期鄞(蛤)药物组>金华药物组>鄞(水洗)药物组>樟村药物组;高剂量组潜伏期鄞(蛤)药物组>金华药物组>樟村药物组>鄞(水洗)药物组;药物组间咳嗽次数比较,无显著差异。5采用呼吸道酚红冲洗法,测小鼠由呼吸道黏膜排出的酚红排泌量,考察四月中上旬于鄞县(水洗;蛤粉)、樟村、金华采收的浙贝母的化痰疗效,结果为:药物组与阴性对照组比较无显著差异(p>0.05)。6采用2,4—二硝基苯酚背部注射制热法,每20min测定大鼠肛温,绘出大鼠3h肛温变化图,考察四月中上旬于鄞县(水洗;蛤粉)、樟村、金华采收的浙贝母的解热疗效,结果为:金华药物组体温变化平缓,能有效抑制发热,效果较好。

【Abstract】 Bulbus Fritillaria Thunbergii is the dried bulbs from perennial herb of Liliaceae. Cold in property, bitter in flavour; meridians entered heart and lung. It can refrigeration and eliminating stagnation, anti-cough and disperse phlegm. Peimine and peiminime are main component, have manyways Pharmacologic Effect.The text has carried on detailed analysis to these respects:1 Character identification and micro-characteristics of Bulbus Fritillaria Medicinal Materials. At the same time, peimine and peiminine were identified by TLC, the results were in line with the requirement of Chinese Pharmacopeia 2005 version.2 The orthogonal design was used, survey the extraction solvent, extraction time, consumption of ammonia liquor , infiltration time of ammonia liquor, to select the optimal extract technic, the best result is: ammonia liquor is 2 times, infiltration 15 minutes, chloroform is 2 times as much as raw materials, accede to soxtherm distil and decocting 4 hours.3 Determination of peimine and peiminine by HPLC, according the result choose the Bulbus Fritillaria Thunbergii to do Pharmacodynamics comparae. Result: harvest at the first and second ten-day period of April from Yinxian, Zhangcun and Jinhua.4 With the method of ammonia caused mice cough, determination of the cough incubation period and degree, survey the effect of anti-cough of the Bulbus Fritillaria Thunbergii which harvest at the first and second ten-day period of April from Yin(washing; clam power), Zhangcun and Jihua. Result: the cough incubation period, the low doses groups were no difference; the middle doses groups Yin(clam power) > Jinhua > Yin(washing) > Zhangcun; the high doses groups Yin(clam power)> Jinhua>Zhangcun> Yin(washing); the cough degree, there was no difference.5 With the method of the excretion quantity of phenofulphthalein in trachea of mice, determination of phenol red output of trachea in mice ,suvrey the effect of disperse phlegm of the Bulbus Fritillaria Thunberigii which harvest at the first and second ten-day period of April from Yin (washing; clam power), Zhangcun and Jinhua. Result: no obvioμs difference between medicines groups and contrast group (p>0.05)。6 With the method of inject 2,4-dinitrophenol in back caused rat heating, determination the rat anus temperature per 20 minutes, draw the change plot of the rat anus temperature in 3 hours,survey the effect of refrigeration of the Bulbus Fritillaria Thunbergii which harvest at the first and second ten-day period of April from Yin(washing; clam power), Zhangcun and Jinhua. Result: the change of the rat anus temperature was slowly in Jinhua group, Jinhua group can effectively inhibit the fever, have good effect.
