化纤胶囊对肝纤维化大鼠防治作用的研究

【摘要】 目的:本研究旨在探讨和揭示肝纤维化的某些发病机理,观察化纤胶囊对肝纤维化的预防和治疗作用。方法:采用改良复合因素法复制肝纤维化大鼠模型,实验动物随机分为空白对照组、模型对照组、秋水仙碱组、化纤胶囊低剂量组、化纤胶囊高剂量组5组。造模各组分别以生理盐水、秋水仙碱悬液、低剂量化纤悬液、高剂量化纤悬液进行干预,测定各组大鼠血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸转移酶(AST)、总胆汁酸(TBA)以及层粘蛋白(LN)、透明质酸(HA)、Ⅲ型前胶原氨端肽(PⅢNP)的含量,同时经HE染色后光镜下观察各组大鼠肝组织形态结构的变化。结果:与空白组比较,模型组大鼠血清中ALT、AST、TBA以及LN、HA、PⅢNP的含量均显著升高(P<0.01),且肝组织形态结构改变最显著;与模型组比较,秋水仙碱组、化纤低、高剂量组血清中ALT、AST、TBA以及LN、HA、PⅢNP的含量均显著降低(P<0.05或P<0.01);与秋水仙碱组比较,化纤低剂量组血清中LN的含量降低更为显著(P<0.05);各治疗组其余各项指标间差异无统计学意义(P>0.05)。秋水仙碱组、化纤低、高剂量组肝组织病变均有不同程度改善,而以化纤低剂量组最为显著。结论:化纤胶囊能显著降低血清中ALT、AST、TBA的含量,表明化纤胶囊有一定程度的保肝降酶的作用;化纤胶囊能显著降低血清LN、PⅢNP、HA的含量,表明化纤胶囊具有改善机体胶原代谢,降低细胞外基质合成和(或)促进细胞外基质降解的作用;化纤胶囊有一定程度的减轻肝细胞变性、坏死,减少肝组织胶原纤维增生,重塑肝组织超微结构的作用,表明化纤胶囊能改善肝纤维化大鼠肝组织损伤程度,对肝纤维化具有一定的防治作用。化纤胶囊与秋水仙碱相比较,其疗效在部分指标上优于秋水仙碱。化纤胶囊低剂量组与高剂量组相比较,低剂量组改善肝纤维化大鼠肝组织损伤程度更显著,表明抗肝纤维化中药治疗用药与病变修复程度具有量效关系,合理剂量用药对已损伤肝功能的恢复具有重要的价值,并已从实验角度证明了肝病治疗用药宜轻清,避免过量伤正以及审因度势、辩证施治、驱邪以扶正的理论价值。

【Abstract】 Objective: To study and explore the pathologic machanism of hepatic fibrosis(HF) and to observe the effect of Huaxian(HX) capsule on the effectiveness of anti-hepatic fibrosis, we made this study.Mathods: The rats models of HF were made by the modified and complexed mathod. The rats were randomized into five groups: black control group, model control group, Colchicine(COL) group, low dosage HX(LHX) group and high dosage HX(HHX) group, and all these groups were treated separately with normol solution(NS), Colchicine, low dosage HX and high dosage HX respectively. Then the content of ALT, AST, TBA, LN, HA and PIIINP in rats’ serums were detected; the section preparations of morphosis changes of rats’ liver tissues were observed under microscope after HE staining.Results: Compared with blank control group,the content of ALT, AST, TBA, LN, HA and PⅢNP in serums of rats of model control group all markedly raised(P<0.01), and morphosis of liver tissues of those were markedly destroyed and changed. Compared with model control group, the content of ALT, AST, TBA, LN, HA and PⅢNP in rats’ serums of LHX group and HHX group markedly reduced(P<0.05 or P<0.01). Compared with COL group, the LN content in serums of rats of LHX group markedly reduced(P<0.05). The other indices among the groups of treatment had no significant differences statistically (P>0.05) .Changes of liver tissue morphosis of rats of COL group, LHX group and HHX group all improved differently and those changes of LHX group improved most markedly.Conclusion: Huaxian capsule can markedly reduce the contents of ALT, AST and TBA in serums of the tested rats. It is suggested that the drug has some influences on the protection of liver and decreasing the contents of enzymes; The capsule can significantly reduce the contents of LN, HA and PⅢNP in serums of rats. It is indicated that the drug can improve collagen metabolism,decrease the synthesis and (or) improve degradation of Extracellular Matrix(ECM); The capsule has the different influence on reducing degeneration and necrosis of liver cells, decreasing hyperplasia of collagenous fibres and remodelling microconstruction of liver tissue. It is shown that the drug has the effect on anti-hepatic fibrosis. Compared with COL, the effect of capsule is superior to that of it in some indices. The effect of LHX group is better than that of HHX group, which suggests that LHX capsule should be used to cure hepatic fibrosis.
