不同穴位处方艾灸催眠作用及其机理的研究

【摘要】 目的:1.观察不同穴位处方艾灸对大鼠戊巴比妥钠致眠时间的影响。2.观察比较三组不同穴位处方艾灸对失眠大鼠下丘脑单胺类神经递质和细胞因子的影响。3.探讨灸法治疗失眠症的作用机理。 方法:1.通过测定戊巴比妥钠致眠大鼠的睡眠持续时间,观察对照组、麦粒灸1组(百会、神道、心俞)、麦粒灸2组(神门、三阴交)和麦粒灸3组(申脉、照海)大鼠的睡眠持续时间的变化。2.以对氯苯丙氨酸(PCPA)所致的失眠大鼠模型为实验对象,通过荧光分光光度测定法,比较对照组、模型组、安定组、麦粒灸1组、麦粒灸2组和麦粒灸3组的大鼠下丘脑5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)、去甲肾上腺素(NE)、多巴胺(DA)含量的变化,通过酶联免疫吸附测定(ELISA)法测定各组大鼠下丘脑白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)的含量变化。 结果:1.三组麦粒灸组与对照组比较睡眠持续时间延长(P<0.05)。2.PCPA腹腔注射后大鼠正常的睡眠-觉醒周期消失,而三组麦粒灸治疗和安定治疗后这一周期均恢复。3.安定治疗后,大鼠下丘脑中5-HT、5-HIAA及NE、DA、IL-1β、TNF-α和IL-6均无明显变化。4.艾灸治疗后,大鼠下丘脑5-HT、5-HIAA含量较模型组明显升高(P<0.05,P<0.01);NE和DA含量较对照组明显降低(P<0.05);IL-1β、TNF-α和IL-6明显升高,与对照组比较差异显著(P<0.05,P<0.01)。5.麦粒灸三组组间,麦粒灸2组明显优于麦粒灸1组(P<0.05),其他组间比较差异无统计学意义(P>0.05)。 结论:1.艾灸能延长大鼠睡眠持续时间,具有镇静催眠作用。2.艾灸治疗与安定治疗均可使PCPA失眠大鼠的睡眠-觉醒周期恢复,但艾灸治疗作用优于安定,灸法可能是通过增加下丘脑IL-1β、TNF-α、IL-6的含量,增加下丘脑5-HT和5-HIAA含量,减少NE和DA含量来治疗失眠症的。3.麦粒灸2组治疗PCPA失眠大鼠优于麦粒灸1组,说明选取神门、三阴交穴位对失眠症的治疗作用可能要优于百会、神道、心俞穴位,为临床选用较好穴位处方治疗失眠症提供了实验依据。

【Abstract】 Objective: 1.To observe the effect of moxibustion on sleep time at different acu-points gruops in sodium pentobarbital rats. 2.To compare the effect of monoamine neurotransmitters and cytokines by moxibustion in insomnia rats’ hypothalamus at different acu-point groups. 3.To explore the mechanism of moxibustion on insomnia.Methods: 1.Twenty-four rats were randomly divided into four groups, control group, grain-sized moxibustion group one (DU20, DU11, BL15), grain-sized moxibustion group two (HT7, SP6) and grain-sized moxibustion group three (BL62, KI6). The different sleep time in these four groups were observed by the method of prolonging sleep time of rat made by sodium pentobarbital. 2.Thirty-six rats were randomly divided into six groups, control group, diazepam group, model group (PCPA), grain-sized moxibustion group one (DU20, DU11, BL15), group two (HT7, SP6), group three (BL62, KI6). Duplicated insomnia model on rats by PCPA abdominal injection, and determined the content of 5-HT, 5-HIAA, NE, DA in hypothalamus with fluorospectropholometry, and the content of TNF-α, IL-1, and IL-6 in hypothalamus with enzyme linked immunsorbent assay (ELISA).Results: 1.The sleep time was significantly prolonged in the three grain-sized moxibustion groups than that of the control group (P<0.05). 2.After PCPA abdominal injection the normal sleep cycle of rats disappeared. Both of grain-sized moxibustion groups and diazepam group could recover the sleep cycle of insomnia rats caused by PCPA. 3.Diazepam has no effect on the content of 5-HT, 5-HIAA, NE, DA , IL-1β, TNF-α and IL-6 in rats’ hypothalamus. 4.After the grain-sized moxibustion treatment, the content of 5-HT and 5-HIAA increased in contrast with those of model group (P<0.05, P<0.01), while that of NE and DA decreased in contrast with those of control group (P<0.05). At the same time, the content of IL-1β, TNF-α and IL-6 increased greatly with comparison to those of control group (P<0.05, P<0.01). 5.In these three grain-sized moxibustion groups, group two has a better effect than group one (P<0.05), but there were not significant differences between the other two groups (P>0.05).Conclusion: 1.Moxibustion therapy can prolong the sleep time, keeping Insomnia rats calm and hypnogogic. 2.
